The Study Of Verification To Characteristic Protein Of Chronic Hepatitis B With Spleen-stomach Dampness-heat Syndrome Based On Drug Intervention

ObjectiveTo verify the expression of differential protein about the pre-selected iTRAQ of CHB with Spleen-stomach dampness-heat syndrome based on drug intervention by Western Blot,observe their changes in the western medicine group,TCM combined with western medicine group and healthy control group,and to obtain the characteristic protein of CHB with Spleen-stomach dampness-heat syndrome based on drug intervention,to explore the regularity of interventional drug and change of expression of characteristic protein with syndrome.For the study of protein molecular markers of the chronic hepatitis B with Spleen-stomach dampness-heat syndrome with biological basis of syndrome,efficacy evaluation and drug targets provide experimental basis.Methods6 cases of each group of the patients of CHB with Spleen-stomach dampness-heat syndrome were collected : the western medicine group,TCM combined with western medicine treatment group,the healthy control group.The peripheral blood was collected in 0,1 and 3 months,the serum was collected and the protein was collected.Using Western Blot technology to detect changes of expression of characteristic protein of pre-selected C1 IN,FETUA,and PLTP between groups,analyzed by mageJ software and GraphPad Prism 5 software.The gender,age and laboratory indexes of each group were analyzed by IBM SPSS Statistics V20.0 software.Results1.The expressions of C1 IN,FETUA,and PLTP were up-regulated before treatment western medicine treatment group and TCM combined with western medicine treatment group compared with the healthy control group of CHB with Spleen-stomach dampness-heat syndrome,and the difference was statistically significant(P<0.05).2.The expressions of C1 IN,FETUA,and PLTP were significantly decreased in the TCM combined with western medicine treatment group comparison before and after treatment of CHB with Spleen-stomach dampness-heat syndrome,the difference was statistically significant(P<0.05).The changes of expression of C1 IN,FETUA,and PLTP in Western medicine treatment group before and after treatment,the difference was not statistically significant.3.Chronic hepatitis B with Spleen-stomach dampness-heat syndrome western medicine group and TCM combined with western medicine treatment group respectively before treatment and after treatment for 3 months,two groups of ALT and HBV-DNA after treatment were lower than those before treatment,the difference was statistically significant(P<0.05),AST and TBIL showed no significant difference;western medicine group and TCM combined with western medicine treatment group after treatment for 3 months,TCM combined with western medicine treatment group HBV-DNA was lower than western medicine treatment group,the difference was statistically significant(P<0.05),and there was no significant difference of ALT.ConclusionThe expressions of FETUA,and PLTP were up-regulated compared with the healthy control group of CHB with Spleen-stomach dampness-heat Syndrom before treatment,which was consistent with the pre-selected result.The level of expression of protein was significantly decreased after intervention with the TCM combined with western medicine treatment group.Consistent with the therapeutic effect of syndrome differentiation and treatment of TCM.It is suggested that it is reliable for the potential therapeutic evaluation markers of the syndrome of CHB with Spleen-stomach dampness-heat syndrome.It is of great significance to study the biological basis of TCM syndrome of CHB,to promote the objectivity of TCM syndrome differentiation.
