ObjectiveLung-Qi exogenous model of rats was used to explore the influence of Shensu Drink on the expression of ?-defensin-2?NLRP3mRNA?Casp1 mRNA and ASC mRNA in lung tissue.Method1 Lung-Qi exogenous model of rats was built by fumigation and injecting lipopolysaccharide(LPS)through caudal veil and blowing cold wind.The changes of weight and the content of IL-6,TNF-? in lung tissue determined with enzyme-linked immunosorbent assay(ELISA)were used to evaluate the model.2 The content of BD-2 in lung tissue homogenate that influenced by Shensu Drink was deternlined with enzyme-linked immunosorbent assay(ELISA).3 The content of NLRP3 m RNA?Casp1 mRNA?ASC mRNA in the lung tissue homogenate was determined with Real-time fluorescent quantitative PCR method(RT-PCR).Result1 The changes of general condition,weight and the content of TNF-?,IL-6 in lung tissue induced by fumigation and injecting LPS through caudal veil and blowing cold wind.The rats appeared less activity and diarrhea.The hairs of rats tarnished and became sparse,yellow.Rales.Their spirits were drooping.The weight of rats in model group grew significantly slowly compared with that of rats in blank group after establishing the rat model for 15 days and 21 days(p<0.01);Compared with the blank group,the content of IL-6 and TNF-a in model group increased significantly(p<0.05).Successfully builded Lung-Qi exogenous model of rats.2.The influence of Shensu Drink on the expression of ?-defensin-2 protein in Lung-Qi exogenous model of rats.Compared with the blank group,The content of?-defensin-2 protein were significantly increased(p<0.05)in the model group;Compared with the model group,the content of ?-defensin-2 protein in lung tissuehomogenate of Shensuyin group were significantly increased(p<0.05);The content of?-defensin-2 protein between Shensuyin high,middle and low group had no significant difference(p>0.05).3.The influence of Shensu Drink on the expression of NLRP3 mRNA ?Caspase1 mRNA?ASC mRNA in Lung-Qi exogenous model of rats.Compared with the blank group,the content of NLRP3mRNA?Casp1 mRNA?ASC mRNA were significantly increased(p<0.05)in the model group;Compared with the model group,the content of NLRP3mRNA?Casp1 mRNA?ASC mRNA in lung tissue homogenate of Shensuyin group were significantly decreased(p<0.05);The content of NLRP3mRNA?Casp1 mRNA?ASC mRNA between Shensuyin high,middle and low group had no significant difference(p>0.05).ConclusionsShensu Drink probably promotes the expression of BD-2 in Lung-Qi exogenous model of rats.Shensuyin may cure Lung-Qi exogenous by up regulating the expression of BD-2.The content of NLRP3mRNA?Casp1 mRNA?ASC mRNA were significantly increased in the model group,indicated that pyroptosis associated with NLRP3 inflammasome may likely participated in the inflammatory process in Lung-Qi exogenous model of rats.Compared with the model group,the content of NLRP3mRNA?Casp1 mRNA?ASC mRNA in lung tissue homogenate of Shensuyin high,middle and low group were significantly decreased,indicated that Shensuyin may could inhibit the course of pyroptosis associated with NLRP3 inflammasome.Shensu Drink promotes the expression of BD-2 in Lung-Qi exogenous model of rats and then inhibits the course of inhibit pyroptosis associated with NLRP3 inflammasome,probably is the target spot in the process of Shensu Drink exerting its efficacy.
