Objective:Lung-Qi exogenous model of mice was used to explore the influence of Shensu Drink on the expression of tissue β-defensin-2in lung and TLR4/NF-κB signal pathway through.Methods:1. The Lung-Qi exogenous model was built by fumigation and injecting lipopolysaccharide (LPS) through caudal veil. The changes of weight, lung index, spleen index and lung in histopathology were used to evaluate the model.2. The content of BD-2in bronchoalveolar lavage fluid that influenced by Shensu Drink was determined with enzyme-linked immunosorbent assay (ELISA).3. The content of proteins in lung that express TLR4, NF-KBp65and BD-2was determined with immunohistochemistry (IHC).4. The content of mRNA in lung that expresses TLR4, NF-KBp65and BD-2was determined with Real-time fluorescent quantitative PCR method.Results:1.The changes of general condition, weight, lung index, spleen index, thymus index of induced by fumigation and injecting LPS through caudal veil. The mice appeared less activity and diarrhea. The hairs of mice tarnished and became sparse, yellow. Their spirits were drooping. The results of histopathological examination in lung showed that the connective tissue of pleura did not proliferate, thicken or damage and there is no inflammation to bronchus in blank group. The connective tissue of pleura proliferated and thickened; inflammatory cells were observed around pulmonary bronchia and vessels with mild nonspecific interstitial pneumonia in model group. The weight of mice in model group grows significantly slowly compared with that of mice in blank group after fumigation for15days (p<0.01); The weight of lung, spleen, thymus of mice in model group were significantly different from that of mice in blank group after fumigation for20days (p<0.05).2. The effect of Shensu Drink on the content of BD-2in bronchoalveolar lavage fluid. The content of BD-2in bronchoalveolar lavage fluid of model group significantly increased compared with that of blank group (p<0.05). The content of BD-2in bronchoalveolar lavage fluid of full formula group significantly decreased compared with that of model group (P<0.05).3. The effect of Shensu Drink on the content of proteins in lung that express BD-2, NF-κBp65and TLR4. The content of proteins that express BD-2, NF-κBp65and TLR4of model group significantly increased compared with that of blank group (p<0.05). The content of proteins that express BD-2, NF-κBp65and TLR4of full formula group, group of benefiting qi for relieving superficies syndrome and group of relieving cough resolving phlegm, significantly decreased compared with that of model group (P<0.05). There is no significant different among whole party group, group of benefiting qi for relieving superficies syndrome and group of relieving cough resolving phlegm.(P>0.05).4. The effect of Shensu Drink on the content of mRNA in lung that expresses BD-2, NF-κBp65and TLR4. The content of mRNA that express BD-2, NF-κBp65and TLR4of model group significantly increased compared with that of blank group (p<0.05). The content of mRNA that express BD-2, NF-κBp65and TLR4of full formula group, group of benefiting qi for relieving superficies syndrome and group of relieving cough resolving phlegm, significantly decreased compared with that of model group (P<0.05). There is no significant different among whole party group, group of benefiting qi for relieving superficies syndrome and group of relieving cough resolving phlegm (P>0.05).Conclusion:BD-2is the target spot of healing Lung-Qi exogenous for Shensue Drink. Shensue Drink probably effects on TLR4/NF-κBp65signaling pathway to promoting the expression of BD-2.
