The Study Of Action Mechanism On Treating Viral Myocarditis With Shensu Drink Modified

Objective:The viral myocarditis(VMC) is causes by many kinds of virus infections,especially Cxoackievirus.The main pathological change is that local or diffuse myocardial cell degenerate,necrosis,infiltrate of non-specificity inflammatory cells in intercellular substance and exude of fibre,what lead to damages of Myocardial, Cardiac dysfunction and(or) Arrhythm- ia.In recent years,the morbidity of viral myocarditis showed an upward trend year after year in our country,and the main sick people are children and young people.But the pathogenesis of VMC has not yet been completely clarified currently,and there was no magic therapy.So the main treatment are rest, symptomatic treatment,providing nutrition for myocardial, anti-oxidation injury and so on.The report of therapy with shensuyin in VMC has not been seen.The purpose of this experiment is seeking more treatments on VMC by studying the therapyeutic effect of the chinese medicine shensu drink modified on the mice that have infected CVB3,which can effectively guide clinic and provide a reliable experimental basis for clinic. Methods:60 male Balb/C mice were divided into 4 groups at random, 15 mice every group,namely:Normal control group(N),Model group(M),Ribavirin group(R),Chinese medicine shensu drink modified group(S). Method of making model:the mice of M group,R group and S group separately inject the virus 10-8TCID50/ml CVB3 Nancy standard liquid 0.1ml in intraperitoneal,and the mice of N group inject normal saline 0.1ml for control.Modeling after an hour,according to conversion formula of dosage of the people and experimental animals,I calculate the dosage of mice is that the mice of R group were given ribavirin 0.2ml/10g (10mg/ml),S group were given Chinese medicin shensu drink 0.2ml/10g (1g/ml),and N and M group were given normal saline 0.2ml/10g.The mice are treatde one time daily and continuous treatment for 10 days. During treatment,I observed the fur,the diet,the behaviour and relieve oneself of every group mice,and weighted them for modeling 3 days ago,modeling day, modeling after three days, model after seven days and putting to death before.All mice were decapitated and the blood were collected and centrifuged for reserve.The left ventricular was removed asepticly and divided into two parts,one part was fixed by 4% formaldehyde solution,and the other was immersed in 70% ethanol solution,what were put in refrigerator 4 degrees for testing.Results:1 Observation of the general conductThe the fur,the diet,the behaviour and relieve oneself of every group mice were normal,and there was no significant difference in the weight of every group mice(P>0.05).During treatment,the mice of every group that were injected CVB3 appeared less activities,aggregation,less diet,less weight,and dull skin, moreover some curly phenomenon.The mice of M group had the death of three,and there was a significant difference in weight of M group contrasting with N group(P<0.01).S and R group separately had a death.Survival mice had better state than M group,and although the weight loss,it was significantly higher than M group(P<0.01).2 The effect of myocardial enzyme with AST and CK with shensu drink modifiedThe content of AST and CK in serum of M group increased significantly,which showed significant difference contrasting with N group(P<0.01).Shensu drink modified and Ribavirin could lower the level of AST and CK in serum,and the two groups had significant difference contrasting with M group(P<0.01). Moreover the level of AST and CK of S group lower than R group,and they had significant difference (AST:P<0.05;CK:P<0.01).3 The effect of SOD activities and the content of MDA in serum with shensu drink modifiedThe SOD activities in serum of M group decreased significantly,which showed obvious difference contrasting with N group(P<0.01).Shensu drink modified and Ribavirin could elevate the SOD activity in serum, and the two groups showed significant difference contrasting with M group (P<0.01). Moreover the level of SOD of S group was higher than R group,and they had difference (P<0.05), the SOD activities of S is higher than R.The content of MDA in serum of M group increased significantly,which showed significant difference contrasting with N group(P<0.01).Shensu drink modified and Ribavirin could decrease the level of MDA in serum, and S group showed significant difference contrasting with M group (P<0.01),R group had significant difference contrasting with M group (P<0.05). Moreover the level of MDA of S group was lower than R group,and they had significant difference (P<0.05).4 The effect of NO in serum with shensu drink modifiedThe level of NO in serum of M group increased apparently, which showed significant difference contrasting with N group (P<0.01).Shensu drink modified and Ribavirin could increase the level of NO in serum.S group had obvious difference contrasting with M group(P<0.01),and R group also showed significant difference(P<0.05).Moreover the content of NO of S group in serum was lower than R group,and they had significant difference(P<0.01).5 The effect of Caspase-3 protein in serum with shensu drink modifiedThe relative content of Caspase-3 protein in myocardial was quantitatively analysed with Fluorescence Index(FI).It was showed that the content of Caspase-3 in M group obviously increased(FI>1.0),which had significant difference contrasting with N group(P<0.01).S group and R group could lower the level of Caspase-3 in myocardial,and showed significant difference contrasting with M group(P<0.01).And the content of Caspase-3 of S group was lower than R group,and they had significant difference(P<0.05).6 The effect of myocardial tissue of VMC mice with shensu drink modifiedThe myocardial of N group had no significant pathological changes.M group,R group and S group had varying degrees of myocardial pathological changes.The myocardial of M group was tuibidity and swelling,cross striation blurred,the eosino- philic staining of cytosolic enhanced,nucleus was pyknosis.It could clearly be seen that many inflammatory cells infiltrate,myocardial cells in large area degenerate and necrosis,myofibril twist and fracture,mesenchmal cells edema. The pathological changes of myocardial of S group significantly reduced,which only showed few inflammatory lesions with lymphocytes.The myocardial of R group with inflammatory cell infiltration and the extent of necrosis was lighter than M group,but seriouser than S group.Conclusion:Shensu drink modified could improve the quality of life of viral myocarditis mice and mitigate the injury of myocardial cell.It could elevate the SOD activity of viral myocarditis mice and lower the content MDA,which had the effect of anti-oxidative damage.It reduced cardiomyocyte apoptosis of viral myocarditis mice.And the curative effect of Shensu drink modified is better than Ribavirin.The study of Action mechanism on treating Viral Myocarditis with Shensu drink modified could provide the basis for chinese clinical treatment of strengthening heslthy qi to eliminate pathotgens.
