Objective:To investigate the expression of Nanog,Lgr5 in cervical carcinoma and its clinical significance.Methods:From 2010 to 2016,a totle of 120 cervix wax specimens were collected from the the gynecology department of the Affiliated Hospital of Jining Medical College,all of which without preoperative radiotherapy and chemotherapy were diagnosed by pathology after surgery.Among them,there are 30 cases of normal cervical tissues,30 cases of cervical intraepithelial neoplasia tissues(10 cases of CINI,12 cases of CINII,8 cases of CINIII),and 60 cases of cervical carcinoma tissues.Using Immunohistochemical SP method,Nanog,Lgr5 was investigated in 60 examples of cervical carcinoma,in 30 examples of cervical intraepithelial neoplasia tissues and in 30 examples of normal cervical tissues.Analysis of the correlation between the positive expression of Nanog and Lgr5,and analyze its relationship with clinicopathological features of cervical cancer.The results of the data were analyzed by SPSS17.0 software package,the percentage(%)was used to count the experimental data,and the comparison of the composition of each group was compared with the x2 test,at the same time,the relationship between Nanog,Lgr5 and the clinicopathological features of cervical cancer was analyzed.The correlation of Nanog and Lgr5 positive expression was analyzed by Spearman correlation coefficient test.Results:1.The positive expression rates of Nanog in cervical cancer tissue,cervical intraepithelial neoplasia and normal cervical tissu e were 76.67%,36.67% and 16.67%respectively,and the difference of positive expression rate was statistically significant(P<0.05).The positive expression rates of Nanog in CIN I,CIN II and CIN III cervical tissues were 20%,33.33% and 62.5% respectively,and the difference of positiveexpression rate was not statistically significant(P > 0.05),but the positive expression of Nanog increased with the increase of CIN grade.The high expression of Nanog was closely related to the degree of tumor differentiation in patients with cervical cancer(P< 0.05),but the relationship between Nanog and the clinical stage,age,pathological type and lymph node metastasis of the patients with cervical cancer was not significant(P>0.05)2.The positive expression rates of Lgr5 in cervical cancer tissue,cervical intraepithelial neoplasia and normal cervical tissue were 73.33%?33.33%?10% respectively,and the difference of positive expression rate was statistically significant(P<0.05).The positive expression rates of Lgr5 in CIN I,CIN II and CIN III cervical tissues were 20%?25%?62.5% respectively.The difference of positive expression rate was not statistically significant(P>0.05),but the positive expression of Lgr5 increased with the increase of CIN grade.The high expression of Lgr5 was closely related to the degree of tumor differentiation d and lymph node metastasis in patients with cervical cancer(P < 0.05),but the relationship between Lgr5 and the clinical stage,age,pathological type of the patients with cervical cancer was not significant(P>0.05).3.The number of positive expressions of Nanog and Lgr5 i n 60 specimens of cervical cancer were 39 cases,Nanog and Lgr5 were negative in 9 cases,Nanog positive expression,and Lgr5 negative expression in 7 cases,Nanog negative expression,and Lgr5 positive expression in 5 cases.According to Spearman test correlation analysis,Nanog and Lgr5 expressions were positively correlated(r= 0.615,P=0.012).Conclusion:1.The expression of Nanog in cervical cancer tissue was enhanced,and had a rising trend in the CIN classification,and was closely related to the degree of tumor differentiation.2.The expression of Lgr5 in cervical cancer tissue was enhanced,and had a rising trend in the CIN classification,and was closely related to the lymph node metastasis and degree of tumor differentiation.3.The expression of Nanog and Lgr5 in cervical cancer tissues was positively correlated,and had a certain synergy in the occurrence and development of cervical cancer.They may have certain value in the evaluation and prognosis monitoring ofcervical cancer.
