Objective:To investigate the expression and clinical significance of Wnt-1,beta-catenin and Lgr5 in cervical cancer.Methods:60 cases of cervical cancer in our hospital from June 2011 to December 2016 were selected and included in the cervical cancer group;30 cases of cervical intraepithelial neoplasia "CIN II~III" cervical conization were included in group HSIL;30 cases of hysteromyoma in our hospital were treated with total hysterectomy in our hospital,and the cervix was confirmed by pathology after operation.The normal group was included in the normal cervical epithelium.The morphological changes of different cervical tissues were observed by HE staining,and the expression of Wnt-1,beta-catenin,Lgr5 in different cervical tissues and the expression of three in cervical cancer tissues were detected by immunohistochemistry and the correlation between the positive rate of the three cases and the different clinical features of cervical cancer was analyzed.Results:1.HE staining results: the cervical epithelial structure was clear,which was divided into the basal layer,the para basal layer,the middle layer and the surface layer.The basal layer cells are arranged vertically,the nucleus of the subbasal layer is larger,the middle layer cells are smaller and dyed deeper than the subbasal nuclei,and there are quite a lot of cytoplasm and "basket knitted form";the surface nuclei are fixed and the nuclear structure disappears.CIN II tissue: mitotic figures mainly exist at the lower 2/3 level of the epithelium,accompanied by pathological mitosis.The proportion of nuclear area in the total area of the epithelial upper layer is about 60-80%,and there is a certain degree of cell polarity.CIN III tissue: the nuclear morphology is abnormal and exists at all levels of the epithelium.The number of mitotic images increased significantly and existed at all levels of the epithelium.The pathological mitotic image existed mostly in the upper layer of the epithelium.The sumof the nucleus area of the nucleus was over 80% in the total area of the cell,and there was no cell polarity.Cervical cancer tissue: the full thickness of the epithelium is occupied by cancer cells,the basement membrane is perforated and infiltrates the stroma.The lesion cells were platelike or nested and had few stroma.The nucleus increased obviously and was polymorphic.2.the results of immunohistochemistry: the positive expression rates of Wnt-l,beta-catenin and LGR5 protein in cervical cancer tissues and CIN II-III tissues were significantly higher than those of normal cervical epithelium(P<0.05).The positive expression rate of Wnt-l,beta-catenin and LGR5 protein in cervical cancer tissues was significantly higher than that of CIN II-III tissue(P<0.05).3.Spearman correlation analysis showed that there was a significant correlation between the expression of Wnt-1 and LGR5,the expression of Wnt-1 and the expression of beta-catenin,the expression of beta-catenin and LGR5 in cervical cancer(P<0.05).4.The expression of Wnt-1,beta-catenin and LGR5 protein in cervical carcinoma increased with the increase of histological grading(P<0.05).Conclusion:1.the expression of Wnt-1,beta-catenin and LGR5 in normal cervical epithelium is low,and is gradually increased in the tissue of CINCIN II-III and cervical cancer.It indicates that the Wnt/ beta-catenin signaling pathway is active in the precancerous lesions and in the occurrence of cervical cancer,which is related to the occurrence and progression of cervical cancer.2.The expression of Wnt-1,beta-catenin and LGR5 has obvious consistency in cervical cancer tissue,and the three has a close correlation.The reason may be related to the target gene of the Wnt signal transduction pathway of LGR5.3.The expression of Wnt-1,beta-catenin and LGR5 increased with the degree of histological grade of cervical cancer,suggesting that the activation of Wnt/ beta-catenin signaling pathway,the target gene LGR5,is involved in the progression of cervical cancer.
