Effect Of Dredging Sanjiao And Qingre Lishi Prescription On Renal TNF-? And Serum IL-2,IL-10 In Rats With IgA Nephropathy

Objective: To observe the effect of dredging Sanjiao and Qingre Lishi Prescription on renal tissue TNF-?,serum IL-2 and IL-10 in rats with IgA nephropathy,and to explore the mechanism of dredging Sanjiao and Qingre Lishi Prescription in the treatment of IgA nephropathy,in order to treat IgA nephropathy clinically.Provide new ideas and methods.Methods: Forty healthy and clean male SD rats weighing 150-180 g were fed for 1 week.The urine protein and occult blood were negative.The weights were randomly divided into 4 groups: normal group(N group)and model group.(Group M),Benazepril Group(Group B),dredging Sanjiao and Qingre Lishi Traditional Chinese Medicine Group(Group Q),10 in each group.The bovine serum albumin + lipopolysaccharide + carbon tetrachloride model was used to replicate the rat model of IgA nephropathy.After the model was successfully replicated,the B and Q groups were given intragastric administration of Benazepril hydrochloride suspension solution,dredging Sanjiao and Qingre Lishi traditional Chinese medicine,and N and M groups were given equal volume of distilled water to gavage once a day for 8 weeks.During the experimental period,the 24-hour urine proteinuria was measured in each group.At the 19 th week,the rats were sacrificed and the serum and kidney tissues were collected to detect blood biochemical indexes,serum IL-2,IL-10 levels,and kidneys.The pathological changes of the globules and the expression of TNF-? in renal tissues.Results:1.The rats in the N group were generally in good condition during the whole experiment;the rest of the rats in the other three groups gradually showed different degrees of apathetic,hairy,lusterless,and reduced feeding.After the corresponding treatment was given,the general state of rats in group B and group Q was improved compared with group M.2.There was no significant difference in liver function and renal function between the 4 groups(P>0.05).3.Proteinuria was not found in the rats of group N during the experiment,and 24-hour urinary protein quantitation was significantly increased in the other three groups.After treatment,group B and group Q were significantly lower than group M(P<0.05).The group and Q group had no statistical significance(P>0.05).4.There was no renal pathological changes in the N group rats,and the degree of pathological changes in the M group was heavier than that in the B group and the Q group(including light microscopy and immunofluorescence).5.The expression of TNF-? in kidney tissue of M group,B group and Q group was significantly higher than that of N group(P<0.05).The expression of TNF-? was significantly decreased in B group and Q group compared with M group(P<0.05).The Q group was no statistically significant(P>0.05).6.Serum IL-2 levels in the M group,B group,and Q group were significantly lower than those in the N group(P<0.05).Serum IL-2 levels in the B and Q groups were significantly higher than those in the M group(P<0.05).Compared with the Q group,no statistical significance(P>0.05).7.The serum levels of IL-10 in group M,group B and group Q were significantly higher than those in group N(P<0.05).The levels of IL-10 in group B and group Q were significantly lower than those in group M(P<0.05);group B,Q The group was no statistically significant(P>0.05).Conclusions:1.Dredging Sanjiao and Qingre Lishi Prescription can significantly reduce 24-hour urinary protein in IgA nephropathy rats,thereby protecting kidney function,and its effect is close to benazepril.2.Dredging Sanjiao and Qingre Lishi Prescription can significantly reduce the pathological damage of kidney tissue in Ig A nephropathy rats and delay the progression of this disease,and its effect is close to benazepril.3.Dredging Sanjiao and Qingre Lishi Prescription can significantly reduce the expression of TNF-? in renal tissue of rats with IgA nephropathy,increase the content of IL-2 in serum and reduce the content of IL-10,reduce the inflammatory reaction,and improve the renal pathological changes.To delay the development of the disease,and its effect is close to benazepril.
