Effect Of Huoxue Tongluo,Qingre Lishi Prescription On Expression Of IL-12,IL-15 And IL-18 In Serum And Renal Tissue Of IgA Nephropathy Rats

Objective: To establish a rat model of IgA nephropathy(IgAN),and to treat IgAN rats with self-made Chinese herbal prescriptions.To detect the expression levels of IL-12,IL-15 and IL-18 in serum and kidney tissues of IgAN rats in different groups,and to explore the correlation between IL-12,IL-15 and IL-18 and the disease,and the possible mechanism of the therapeutic effect of self-made Chinese herbal prescriptions on the disease.Methods: The experimental rats were randomly divided into normal group(N group),model group(M group),western medicine group(X group)and traditional Chinese medicine group(Z group).IgAN rat model was established by referring to literature.After the model was replicated successfully,Benazepril hydrochloride was given to group X,Huoxue Tongluo,Qingre Lishi Prescription was given to group Z,and distilled water was given to group N and M daily for 4 weeks.During the experiment,24-hour urinary protein(24h-UTP)was detected in rats by stages.At the 14 th week,blood was collected,blood biochemistry was detected,and kidney tissue samples were collected for RT-PCR,light microscopy and immunofluorescence detection.Results:1.The mental state,hair and diet of rats in group N were normal;the general situation of rats in the other three groups gradually deteriorated during the modeling period;after drug intervention,the general situation of rats in group X and group Z was better than that in group M.2.Before modeling,24h-UTP in rats was negative;after modeling,24h-UTP in M,X and Z groups was higher than that in N group(P<0.05);after treatment,24h-UTP in X and Z groups was lower than that in M group(P<0.05).3.There was no significant difference in serum creatinine,urea nitrogen,albumin,triglyceride and low density lipoprotein among the groups(P>0.05).4.Immunofluorescence: There was no change in kidney tissue in group N;immunocomplex deposition was observed in group M,group X and group Z;compared with group M,immunocomplex deposition was decreased in group X and group Z after drug treatment.Light microscopy: The morphology and structure of kidney tissue in group N were not changed;the pathological changes of kidney in group M were more serious;compared with group M,the pathological changes of kidney tissue in group X and Z were alleviated after drug treatment.5.The expression of IL-12,IL-15 and IL-18 in kidney tissue was observed by RT-PCR.Compared with group N,the expression of IL-12,IL-15 and IL-18 was increased in group M(P<0.05),but decreased in group X and group Z(P<0.05).6.The levels of IL-12,IL-15 and IL-18 in serum of IgA nephropathy rats were observed by ELISA.Compared with group N,the levels of IL-12,IL-15 and IL-18 in group M increased(P<0.05),while those in group X and group Z decreased(P<0.05).Conclusion:Huoxue Tongluo,Qingre Lishi Prescription could reduce 24-hour urinary protein content in IgA nephropathy rats.It can alleviate the pathological damage of kidney tissue in rats with IgA nephropathy,reduce the contents of IL-12,IL-15 and IL-18 in serum of rats with IgA nephropathy,and decrease the expression of IL-12,IL-15 and IL-18 in kidney tissue.
