Establishment Of Xenograft Model Of Renal Cell Carcinoma Model In Nude Mice And The Study Of The Effect Of PNS Assisting Sunitinib And IFN-α2b On The Xenograft Model

The incidence of renal cell carcinoma which is not sensitive to conventional radiotherapy and chemotherapy is increasing year by year in our country,such as IFN-α2b.The effect of immunotherapy is very limited,meanwhile,probably accompanied by serious toxic and side effects,which hinders the wide application of immunotherapy in clinic.Most of the patients when diagnosed with renal cell carcinoma have advanced to the late stage.About 17% of the patients have distant metastasis causing not obvious curative effect of radical surgery and the prognosis of patients is generally poor.In recent years,with the deepening of research on the molecular mechanisms of renal cell carcinoma,produce some specific targeted therapies,such as sunitinib and bevacizumab.The emergence of molecular targeted therapy greatly improved the prognosis of patients with renal cell carcinoma,significantly prolonging the Progression Free Survival(Progression Free Survival,PFS)and the Overall Survival(Overall Survival,OS).However,drug resistance is liablely generated with targeted drugs,which seriously affects the treatment effect in the later period of the patients.Chinese herbal medicine has been a good recipe for the treatment of various diseases since ancient times.Now its application in cancer has become a hot spot of research.Panax notoginseng belongs to Araliaceae,with functions of dispeling stasis and stoping bleeding,swelling and pain,commonly used in hemoptysis,hematemesis,epistaxis,hematochezia,uterine bleeding,traumatic bleeding and other symptoms.Total saponins of panax notognseng is the main chemical components of Panax notoginseng,containing more than 60 kinds of monomeric saponins,including ginsenoside Rb 1,Rg1,Rg3,Rc and so on.Since 1940 s,Domestic and foreign scientists have launched a series of researches on PNS,revealing the role of PNS in central nervous system,cerebrovascular system,cardiovascular system,hematopoietic system and immune system.In recent years,the research of PNS in tumor and its related complications has also become a hot spot.This study found that total saponins of panax notognseng could inhibit tumor growth in nude mice that is the xenografted model of renal cell carcinoma,improve immunity,weakening the side effects of targeted drug sunitinib and increasing drug sensitivity of sunitinib.The possible molecular mechanisms may involve SRE,SBAP1,HSF and RARE.Therefore.PNS is likely to be a new and effective drug for the treatment of kidney cancer.
