Objective:Establish the subconjunctival hemorrhage model in rabbit eyes,Set up the efficacy indicators and quantitative indicators of subconjunctival hemorrhage, Discuss the pharmacological effects of notoginseng total saponin (PNS), Confirmed notoginseng total saponin eye drops is feasible and effective, in the treatment of subconjunctival hemorrhage in rabbit eyes.Method:In the experiment,20healthy adult New Zealand white rabbit (40eyes) were divided into treatment group and blank control group, Among them, according to the different drug concentrations,Treatment group is divided into high concentration group, middle concentration group, low concentration group, each group has5(10eyes).By injecting autologous venous blood under conjunctiva, To establish a model of subconjunctival hemorrhage. After modeling, Treatment group was given notoginseng total saponin eye drops:the low concentration group is0.5%, middle concentration group is1%, high concentration group is2%, Drugs withheld until the subconjunctival hemorrhage was cured. Blank control group not given any treatment. After modeling, we observed the change of the subconjunctival hemorrhage under the slit lamp microscope,and then record the heal time.Set up the curative effect index:5days to cure is significant,6-7days to cure is effective, after7days did not cure is invalid, Statistics the number of cases in each group,Then using SPSS17.0to K-W inspection and U inspection.After Modeling1d,3d,5d,7d,9d, we take conjunctival photos (the amplification of16times)of each sample, by slit lamp microscope, Using Image-Pro Plus to measure area, mean optical density (MOD), integrated optical density (IOD) of subconjunctival hemorrhage region,Then we variance analysis of repetitive measure by SPSS17.0. By two methods, we can measure the treatment effect of notoginseng total saponin.Results:1.According the heal time to measure:The average healing time of treatment groups (low concentration, middle concentration group, high concentration group) is less than the blank control group, Its total effective rate is significantly higher than the blank control group, And it is proportional to concentration. The curative effect exist significant differences between groups (P<0.05). Pairwise comparison results display:between treatment group and blank control group,the curative effect was significant difference,the differences are significant between low concentration group and high concentration groups,but the low concentration group compared to the middle concentration group,the middle concentration group compared to the high concentration group, there was not significant difference.2. According the area to measure:The longer the treatment time, the smaller the bleeding area. The high concentration group reduce fastest. Area values exist significant differences between groups (P<0.05), Pairwise comparison results display:treatment groups compared to the blank control group, there was significant difference,(P<0.05), But no significant difference among treatment groups (low concentration, middle concentration group, high concentration group)(P>0.05)3. According the mean optical density (MOD) to measure:The longer the treatment time, the smaller the mean optical density. The high concentration group reduce fastest.MOD exist significant differences between groups (P<0.05), Pairwise comparison results display:Multiple comparisons between groups, MOD was significantly different (P<0.05), expect the low concentration group compared to the blank control group, the middle concentration group compared to the high con-centration group (P>0.05)4. According the integrated optical density (IOD) to measure:The longer the treatment time, the smaller the integrated optical density. And the high concentration group reduce fastest. IOD exist significant differences between groups (P<0.05), Pairwise comparison results display:Multiple comparisons between groups, IOD was significantly different (P<0.05), expect the low concentration group compared to the middle concentration group and the blank control group, the middle concentration group compared to the high concentration group (P>0.05)Conclusion:1. By injecting autologous venous blood under conjunctiva, we can establish the model of subconjunctival hemorrhage. It make the model unified, standardize experimental treatment factors.2. PNS eye drops was used for the treatment of subconjunctival hemorrhage in rabbit eyes. Compare the heal time, Area, MOD and IOD. Confirmed that PNS eye drops can accelerate the absorption of subconjunctival hemorrhage Make the bleeding area reduce significantly, the bleeding color becomes shallow, Shorten the time of the disease. And with the drug concentration, curative effect increased.3. It is more objective and effective to use IPP measure Area, MOD, IOD. While IOD includes Area and MOD, It is the best indicators of subconjunctival hemorrhage.
