Objective:To observe the IL-1?content in periodontal tissue and peripheral blood of rats with periodontitis and to explore the functional mechanism of BUSHENHUOXUEGUCHIFANG in treating periodontitis.Method:3-month-old male SD rats were randomly divided into three groups:blank control group,model group,treatment group.Model:After anesthesiathe,the left maxillary first molar of rats in the model group and treatment group was ligated with 0.2 mm diameter orthodontic stainless steel ligation wire,and gingiva was stripped.After that,the rats were fed with high sugar diet and high sugar water by 4 weeks[1].Rats in the blank control group on the anesthesia day were fed with routine feed and water,without periodontal treatment[2].After the model was established successfully,the ligation were removed from the experimental teeth in the model group and the treatment group rats under anaesthesia.On the same day,the blank control group rats were given the same anesthesia without periodontal treatment.Fromthenextday,thetreatmentgroupwasgiven BUSHENHUOXUEGUCHIFANG for 4 weeks,and the model group and the blank control group were given the same dose of normal saline for 4 weeks.After 4 weeks of gavage,IL-1?content in gingival tissues around the experimental teeth and in peripheral blood of 3 groups of rats was detected by Real-time PCR method and enzyme linked immunosorbent assay?ELISA?,respectively,and the data were analyzed by SPSS21.0 software F test and SNK test.Results:After 4 weeks of modeling,the gingival around the experimental teeth are red and swollen,soft and bleeding easily in the model group and the treatment group rats.Slice showed that the gingival was infiltrated by a large number of inflammatory cells,capillary dilated,junctional epithelium root proliferated to form a periodontal bag,and the periodontal ligament space was widened,osteoclasts were seen in the alveolar bone,and alveolar bone resorpted.There was no obvious inflammation and uneasy to bleed in gingiva of rats in the blank control group,and the slice showed that the periodontal tissue structure was intact,and there was no inflammatory cell infiltration in gingival tissue,no bone resorption in alveolar bone and rootless proliferation and displacement in junctional epithelium.After 4 weeks of gavage,there was no obvious inflammation and uneasy to bleed in the gingiva of experimental teeth in the control group and the treatment group.The gingival texture of rats in the model group was red and swollen,soft and bleeding easily.The results showed that the periodontal condition in the treatment group was improved.The IL-1?content of gingival tissues in the treatment group was lower than that in the model group?P<0.05?,but there was no significant difference between treatment group and blank control group?P>0.05?.The IL-1?content of gingival tissues in the model group was higher than that in the blank control group?P<0.05?.The IL-1?content of peripheral blood in the treatment group was lower than that in the model group?P<0.05?,but was higher that that in the blank control group?P<0.05?.The IL-1?content of peripheral blood in the model group was higher than that in the blank control group?P<0.05?.Conclusions:1.The animal periodontitis model can be successfully built through applying periodontal ligation and stripping gingiva while feeding sticky high-sugar diet.2.The BUSHENHUOXUEGUCHIFANG is effective in lowering IL-1?in periodontal tissues and peripheral blood.3.The BUSHENHUOXUEGUCHIFANG can treat periodontitis by reducing the IL-1?in periodontal tissues and peripheral blood.
