Objective: The purpose of the study is to explore the effect of ginkog biloba extract(GBE)on periodontitis by constructing experimental periodontitis model in SD rats,which will provide relevant basis on the treatment of periodontitis by GBE.Methods:1.The effect of GBE on experimental periodontitis in SD rats: 24 Specific-pathogen-free male SD rats at the age of 5 weeks were randomly divided into 4 groups(group A: normal control group;Group B: periodontitis model group;Group C: GBE low dose group: 50mg/kg;Group D: GBE high dose group: 300 mg/kg).After adaptively feeding 2 weeks,the rats in B,C and D group were set up periodontitis model of 4-0silk thread ligated bilateral maxillary second molars.2.The effect of ginkgo biloba extract on periodontitis: after 4-0 thread ligation bilateral maxillary second molar for 8 weeks,A and B group were gaven distilled water by gavage(2 ml/d);C and D group were respectively given GBE in low dose(50 mg/kg,2 ml/d)and GBE in high dose(300 mg/kg,2 ml/d)for five weeks.After 24 h in the last gavage,all the rats(group A-D)were killed by taking off the neck.Weight changes were recorded as well as the samples on one side were collected to detect the CEJ-AC distance and the samples on the other side were performed HE staining to observe the histological changes.Results:1.Before the experiment,the weight of rats in group A-D was 193±16 g,203±12 g,192±14 g and 209±12 g,respectively.There was no statistically significant difference among them(P>0.05).After the ligation of teeth for 2 weeks,only was the weight of rats in the periodontitis group significantly decreased(**P<0.01,compared with the normal group),while that of the other groups was not significantly changed(P>0.05,compared with the normal group).Subsequently,the weight of periodontitis group was lower than normal group(10,11 w **P<0.01,12 w *P<0.05,13-20 w **P<0.01).The weight of GBE low dose group were slightly lower than normal group,and after GBE gavage,there were no significant changes compared with the normal group(10 w **P<0.01,11,12 w **P<0.01,13 w**P<0.01,14 w P<0.05,16 w **P<0.01,15,17-20 w P>0.05).The weight in GBE high dose group were no different than normal group(P>0.05).2.After GBE gavage for 5 weeks,the CEJ-AC distances in four groups were 0.36±0.08mm(the normal group),0.78±0.22 mm(periodontitis group),0.50±0.17 mm(GBE low dose group)and 0.67±0.17 mm(GBE high dose group).Compared with the normal group,the CEJ-AC distances of other three group increased,and the difference were statistically significant(**P<0.01).Compared with periodontitis group,the CEJ-AC distances were reduced in GBE low dose group and GBE high does group.But only in GBE low dose group,the difference was statistically significant(#P<0.01).Compared between two GBE gavage groups,the CEJ-AC distance in GBE low dose group was smaller than that of GBE high dose group.The difference was statistically significant(?P<0.01).3.HE staining results showed that: compared with normal group,the gum normal morphology and structure in periodontitis group disappeared.The normal morphological structure of gingival tissue in the GBE low dose group partially disappeared,while the normal morphological structure of gingival tissue in the GBE high dose group mostly disappeared.Conclusion:1.GBE can effectively reduce the alveolar bone absorption of SD rats with experimental periodontitis.2.The GBE low dose(50 mg/kg)may have a better effect than GBE high dose(300mg/kg)on the treatment of experimental periodontitis in SD rats.
