Objective: To investigate the expression of progesterone-induced blocking factor(PIBF),interleukin-2(IL-2)and interleukin-10(IL-10)in venous blood of patients with threatened abortion,To investigate the effect of PIBF on Th 1 / Th2 balance in normal pregnancy and to study the mechanism of PIBF in aura(recurrent)abortion.Methods: The study group was divided into study group and control group.The study group consisted of 40 patients with threatened abortion who were divided into progesterone group and dydrogesterone group.The dydrogesterone group was treated with dydrogesterone in 20 cases,The progesterone group was treated with progesterone in 20 cases.The control group included 40 women with the same pregnancy status during normal pregnancy.The expression of P,PIBF,IL-2 and IL-10 in the venous blood of the study group and the control group were detected by enzyme-linked immunosorbent assay(ELISA).The Wilcoxon sign and t test were used to analyze the differences between the study group and the control group.Spearman correlation analysis was used to analyze the correlation between the expression levels of PIBF,IL-2 and IL-10,and the difference was statistically analyzed.Results:1.In the study group before treatment,serum P(53.47±2.15)ng/ml,PIBF(129.27±4.48)pg/ml,IL-2(260.00±7.95)ng/L,IL-10(282.74±8.02)ng/L,the control group before treatment serum P(65.25±1.28)ng/ml,PIBF(175.26±4.92)pg/ml,IL-2(225.00±6.29)ng/L,IL-10(319.28±7.19)ng/L.In the study group,the levels of PIBF and IL-10 in the venous blood were lower than those in the control group,IL-2 was higher than that in the control group,the difference was statistically significant(P <0.05).2.In the study group after treatment,serum P(81.26±4.65)ng/ml,PIBF(135.35±5.08)pg/ml,IL-2(146.35±6.65)ng/L,IL-10(381.60±8.52)ng/L,P?PIBF ?IL-10 was higher than that in the study group before treatment,IL-2was lower than that in the study group before treatment.The difference was significant(P <0.05).After treatment with progesterone,Serum P(77.49±4.47)ng/ml,PIBF(128.68±5.09)pg/ml,IL-2(127.26±5.91)ng/L,IL-10(342.65±7.49)ng/L,Compared with pre-treatment,P?PIBF ?IL-10 was higher than that in the progesterone group before treatment,IL-2was lower than that in the progesterone group before treatment.The difference was significant(P <0.05).After treatment with progesterone,The levels of serum P(89.27±5.82)ng/ml,PIBF(162.36±7.08)pg/ml,IL-2(158.26±2.16)ng/L,IL-10(439.13±8.66)ng/L,Compared with pre-treatment,P?PIBF ?IL-10 was higher than that in the progesterone group before treatment,IL-2was lower than that in the progesterone group before treatment.The difference was statistically ignificant(P <0.05).The expression of PIBF and IL-10 in the progesterone group was significantly higher than that in the progesterone group,while the IL-2 level was significantly decreased(P <0.05).3.The study group was negatively correlated with PIBF and IL-2(r=0.421,P<0.05),and IL-10 was positively correlated(P <0.05).There was a positive correlation between progesterone level and PIBF level before and after treatment(P <0.05).But there was no correlation between progesterone level and PIBF level in the study group before and after treatment(r=0.128,P>0.05;r=0.157,P>0.05).4.There was a significant difference between the two groups(P> 0.05).The effect of dibenzoin on PIBF was stronger than that of progesterone.Conclusions:1.Downregulation of PIBF expression may affect the Th1 / Th2 equilibrium state to the Th1 cytokine shift.The level of PIBF was up-regulated significantly in patients with threatened abortion after appropriate progesterone maternal treatment,Th1 / Th2 balance state to Th2 cytokine shift significantly,indicating that progesterone can effectively reduce the possibility of threatened abortion..2.Through impact the level of PIBF of pregnant women,Progesterone can protect the embryo,to some extent,the possibility of reduction of threatened abortion.Detection of PIBF levels in pregnant women during early pregnancy can be used to assess the therapeutic effect of progesterone and the risk of threatened abortion.
