| Abortion refers to those who are less than 28 weeks pregnant and whose fetus weighs less than 1 kg and terminates.Among them,abortion caused by natural factors is spontaneous abortion,threatened abortion is the first stage of the development of spontaneous abortion.In recent years,with the opening of the national "two-child" policy,the development of assisted reproductive technology,the increase in social pressure and changes in living habits,the situation of spontaneous abortion has become increasingly severe.Therefore,how to improve pregnancy outcomes is still the focus of current research on spontaneous abortion.This study first explored the fetal protection effect of different doses of ramie root decoction on the mifepristone induced abortion model in rats through the method of traditional Chinese medicine pharmacology,and explored the regulatory effect of ramie root on Th1 / Th2 / Th17 / Treg balance from the perspective of reproductive immunology.Sexually mature SD(Sprague-Dawley)rats were mated with males and females to establish a pregnant rat model.The pregnant rats were divided into a normal group,a model group,a drug administration group(low,intermediate,and high-dose ramie root)and progesterone.From the first day of pregnancy,the administration group was given the corresponding drugs,the normal group and the model group was respectively given an equal amount of saline.On the 10 th day of pregnancy,except for the normal group,the other five groups were administered with mifepristone 3.75 mg / kg by gavage to establish an abortion model.Pregnant rats were sacrificed at 16 days of pregnancy to obtain materials such as blood and placenta.1.Count the number of implants,live babies,fetal retention rate and uterine fetal weight in each group of pregnant rats;2.Use enzyme-linked immunosorbent assay(ELISA)to detect progesterone(P),Chorionic gonadotropin(β-HCG),Th1-(IFN-γ),Th2-(IL-4),Th17-(IL-17A),and Treg-(TGF-β1)levels;The levels of transcription factors Th1-(T-bet),Th2-(GATA-3),Th17-(ROR-γt)and Treg-(Foxp3)in peripheral blood and placental tissues of pregnant rats were detected by PCR.The research results are as follows:1.The number of implantation of pregnant rats in each group was basically the same.Compared with the model group,embryo protection rate and uterine continuous fetal weight of the ramie root medium-dose group increased significantly;2.Compared with the model group,the serum P and β-HCG levels of pregnant rats in the middle dose group of ramie root increased significantly,and the levels of cytokines and transcription factor genes related to Th2 and Treg cells in peripheral blood were significantly increased,while those of Th1 and Th17 cells were significantly decreased.3.In the ramie root medium-dose group,compared with the aborted placental tissue,the normal placental tissue not only showed morphological changes,but also showed a significant increase in P and β-HCG levels.In summary,the results of this study indicate: 1.The decoction of ramie root decoction in each dose group basically does not affect the implantation of SD pregnant rats,and the ramie root medium dose group has a significant fetal preservation effect.It may play a role in protecting the fetus by increasing the levels of P and β-HCG in the serum and placenta of pregnant rats to regulate the Th1 / Th2 / Th17 / Treg balance in the peripheral blood and placental tissue of aborted pregnant rats.Then through the pharmacology of the Chinese Medicine Network to predict the pharmacological substance basis of ramie root fetal protection,related targets for fetal protection and the signal pathways involved in fetal protection,to further understand the role of ramie root for fetal protection and provides a certain reference value for further research the action mechanism on ramie root for fetal protection.In order to preliminary explore the medicinal ingredients and mechanism of ramie root in the treatment of spontaneous abortion,the chemical constituents of ramie root were collected firstly by using TCMSP,chemical database,and so on.And the active ingredients of ramie root were selected according to pharmacokinetic parameters,subsequently predicted the potential protein targets of ramie root.At the same time,used DiGSeE and other searches to search for genes related to natural abortion,and matched the ramie root-related targets with abortion-related targets to obtain potential targets for ramie root active ingredients to treat abortion,and the pathways were enriched and analyzed for these targets,finally the active components-targets-pathways network of ramie root was visualized.The research results are as follows:1.The levels of cytokines,transcription factor proteins and their genes associated with Th2 and Treg cells were significantly increased,while the levels of cytokines,transcription factor proteins and their genes associated with Th1 and Th17 cells were significantly reduced.2.A total of five active ingredients were screened out for ramie root,including: quercetin,(+)-catechin,mairin,β-sitosterol,and hederagenin.3.Ramie root for spontaneous abortion had 18 potential targets,of which there are 7 signal pathways involved,namely the AGE-RAGE signaling pathway in diabetes complications,malaria,intestinal immune network for IgA generation,HIF-1 signaling Pathways,allograft rejection,complement and coagulation cascades,and the NF-κB signaling pathway.The research has preliminary predicted the mechanism of ramie root in the treatment of spontaneous abortion,which provides clues for further in-depth study of ramie root’s fetal preservation effect.The above results indicate: Quercetin has an important role in fetus protection of ramie root,but it is not this single component that plays a role in fetal protection alone;ramie root treatment of abortion is mainly carried out through immune,coagulation,apoptosis,and nervous system regulation. |
