Objectives To study PHD2 as HIF-1? upstream regulatory genes may be related to the occurrence of cervical cancer development,and study the expression significance of the high-risk type HPV infection,PHD2,HIF-1?,P53 in different cervical lesion and the correlation.To provide scientific theoretical basis for the diagnosis and prevention of cervical cancer,and to provide a reference for its prognosis in clinic.Methods From January 2016 to August 2017 department of wax block,cervical intraepithelial neoplasia stage I(CINI group,20 cases),cervical intraepithelial neoplasia II and III(CIN2-3 group,20 cases)and cervical cancer(30 cases)according to the pathological classification into corresponding to the four groups of 14 kinds of high-risk HPV DNA types.Gather specimens of patients with pathologic and clinical data,for using the fluorescent quantitative PCR technique to detect 14 kinds of high-risk HPV DNA points,including HPV16,18 and other 12 kinds of high-risk HPV subtypes(31,33,35,39,45,51,52,56,58,59,68,68).Immunohistochemical method was used to detect the expression levels of PHD2,HIF-1?and P53 in each group.Analyze the relationship between PHD2,HIF-1? and P53 expression in cervical carcinoma with pathological grading,clinical staging,and the Correlation between PHD2,HIF-1?,P53 and HR-HPV.The differences of expression of PHD2,HIF-1?,P53 between 4 groups was analyzed by OneWay ANOVA single factor variance analysis.The difference in the positive rate between the experimental group and the control group was examined by partitions of ?2 method.The differences of expression of PHD2,HIF-1?,P53 in different pathological grades and clinical stages were analyzed by chi-square test and Fisher's exact probability.The relationship between the expression of PHD2,HIF-1?,P53 and HR-HPV was analyzed by Spearman correlation.Results 1 The positive detection rates of PHD2 in normal cervical tissues,CIN1,CIN2-3 and cervical cancer were 5%,30%,45% and 80% respectively.HIF-1? positive expression rate were 10%,40%,55%,86.6%,P53 positive expression rate were 10%,30%,40% and 70% respectively.The expression levels of PHD2,HIF-1? and P53 in different cervical tissues were different(PHD2: F = 32.610,P <0.001;HIF-1? :F = 38.495,P < 0.001;P53: F = 17.790,P< 0.001).The positive expression rate of PHD2,HIF-1? and P53 was not statistically significant in normal cervical tissue and CIN1,The positive expression rate of P53 was not statistically significant in normal cervical tissue and CIN2-3,The positive expression rate of PHD2,HIF-1? was statistically significant in normal cervical tissue and CIN2-3,the positive expression rate of PHD2,HIF-1? and P53 was statistically significant in normal cervical tissue and cervical cancer tissue.2 In the relationship of clinicopathological features of cervical cancer patients,it is related to the degree of differentiation(PHD2:?2=11.905,P=0.001;HIF-1?:?2=12.269,P=0.003;P53:?2=10.169,P=0.001),clinical stage(PHD2:?2=8.268,P=0.005;HIF1?:?2=5.099,P=0.037;P53:?2=4.833,P=0.032)and lymph node metastasis(PHD2:?2=5.762,P=0.026;HIF-1?:?2=6.692,P=0.018;P53:?2=12.377,P=0.001),regardless of age(PHD2:?2=3.625,P=0.057;HIF-1?:?2=2.231,P=0.272;P53:?2=2.762,P=0.097).3 Correlation analysis showed that: PHD2,HIF-1? and P53 were positively correlated with high-risk HPV positive expression.The expression of PHD2,HIF-1?and P53 ofthe hypoxia induction factor signal transduction pathway in cervical cancer was positively correlated with.Conclusions 1 It is suggested that PHD2,HIF-1?and P53 are highly expressed in cervicalcancer tissues and play an important role in the development of cervical cancer.2 The positive expression rate of HPV in cervical intraepithelial neoplasia and cervical cancer is more than 90%,and the incidence and mortality of cervical cancer can reduce by preventing and controlling the infection rate of high-risk HPV.3 The positive expression rates of PHD2,HIF-1? and P53 in cervical cancer tissues and their clinicopathological features were correlated with pathologic differentiation,lymph node metastasis and clinical staging,but were not related to age.4 The malignant progression of PHD2,HIF-1? and P53 in cervical cancer is expected to be a new target for cervical cancer targeting therapy.There is a positive correlation between PHD2,HIF-1? and P53 and positive expression of high-risk HPV,and may contribute to the progression of cervical lesions by mutual interaction.
