Purpose:To investigate the expression level of PHD2 in ovarian cancer and its effect on the migration,proliferation and platinum resistance of ovarian cancer cells.Experimental Design:The expression of PHD2 in the tissues of the patients with ovarian cancer and the control group(normal ovary and cervical cancer)were detected by RT-PCR,immunohistochemistry and Western Blot.The PHD2 level was transfected by si RNA via lipofectamin2000 in ovarian cancer cell lines.The migration ability of the cell was detected by Wound Healing.We detected the proliferation ability and the change of the cisplatin sensitivity via CCK8.The co-culture system was constructed by inserting cell culture dish Millicell to explore the effect of PHD2 and CAFs on ovarian cancer cells.Results: The expression of PHD2 in ovarian cancer was higher than that of normal ovary and cervical cancer.After knocking down PHD2 in SKOV3 cell line,TGF-?1 was down synchronously and the migration ability of SKOV3 decreased.The proliferation ability was not significantly changed.The sensitivity to platinum drugs increased and IC50 decreased.When SKOV3 was co-cultured with CAFs,the proliferation ability of SKOV3 increased.In the co-culture system,knockdown of PHD2 in the CAFs caused the decrease of SKOV3 proliferation and migration ability.Conclusions:PHD2 is highly expressed in ovarian cancer,which promotes the migration and platinum resistance of ovarian cancer through the TGF-?1 pathway.The expression of PHD2 in CAFs affects the development of ovarian cancer.
