Objectives By thyroid extirpation combined propranolol lavage method to establish the heart Yang deficiency and brady arrhythmia model in rats,to observe the effect of Lvshe-ng Xinkang on the anus temperature,heart rat,heart Yang deficiency scale score,histopathological changes in heart tissues,Cx45,Gs?,HCN4,cAMP,cGMP,cAMP/cGMPof heart Yang deficiency syndrome and brady arrhythmia in rats,discussion its mechanism of action,exploration the effects and mechanisms of Lvsheng Xinkang for further clinical treatment.Methods Selested SD rats,and adaptive feed for a week.10 rats were randomly selected as the normal group,the others to establish heart Yang deficiency model by thyroidectomy.The heart Yang deficiencied rats were randomly divided into four groups,for the model group,xinbao group,Lvsheng Xinkang high group,and Lvsheng Xinkang low group,after the model of heart Yang deficiency was prepared successfully.Every morning,the normal group was given physiological saline gavage,the other groups were given propranolol 15mg/kg gavage.Every afternoon,the normal group and the model group was given physiological saline gavage,the Xinbao group was given Xinbao pil gavage,the Lvsheng Xinkang highgroup and Lvsheng Xinkang low group were given corresponding Lvsheng Xinkang decoction,for 7 days.Observation the changes of the rats' anus temperature,heart rat,heart Yang deficiency scale score,the pathological and histological changes in myocardical by HE staining.Using immunohistochemical method measuring the rat' CX45.Using Western Blot method measuring the rat' Gs? and HCN4.Using ELISA method measuring the rat' c AMP,cGMP,cAMP/cGMP.Evaluation the model results and the impact of Lvsheng Xinkang on Cx45,Gs?,HCN4,cAMP,cGMP and cAMP/cGMP.Results 1Compared with the normal group,the model group rats of anus temperature,heart rate significantly decreased(P<0.05);Compared with the model group,the Lvsheng Xinkang group rats of anus temperature,heart rate significantly increased(P<0.05).2Com-pared with the normal group,the model group rats in bad condition,rough coat,spiritual malaise,slow response,reduced active,thin and small,large toilet thin,cold tail,the degree of heart Yang deficiency in model group was significantly increased(P<0.05);Compared with the model group,the Lvsheng Xinkang group rats' degree of heart yang deficiency was significantly decreased(P<0.05).3Compared with the normal group,the model group rats of the myocardial fiber swelling and disorder with a wave like change and some myocardial fibers defected as well as fractured,nucleolus shrinked and rounded could be coloured with an uneven distribution.myocardial cell degeneration and swell;Compared with the model group,the Lvsheng Xinkang group were improved to some degree.4Compared with the normal group,model group Cx45,Gs? and HCN4 significantly decreased;Compared with the model group,the Lvsheng Xinkang high group and Lvsheng Xinkang low group Cx45,Gs? and HCN4 significantly increased(P<0.01).5Compared with the normal group,the model group cyclicAMP content decreased(P<0.05),cyclicGMP content increased(P< 0.05),cAMP/cGMP ratio decreased(P<0.05);Compared with the model group,the Lvsheng Xinkang high group and Lvsheng Xinkang low group cyclicAMP content increased(P< 0.05),cyclicGMP content decreased(P<0.05),cAMP/cGMP ratio increased(P<0.05).Conclusions 1Lvsheng Xinkang can improve the rat anus temperature,heart rat,increase the content of cAMP,reduce the the content of cGMP,increase the cAMP/cGMP ratio,make the heart Yang deficiency syndrome to improve,with statistical significance.2Lvsheng Xinkang has therapeutic effect on heart Yang deficiency typed brady arrhythmia,which may be related to improve myocardial cell structu and increase the expression of Cx45,Gs? and HCN4.
