Objective:Observe the effect of xixin and fuzi with different proportions on B rady arrhythmia (Ba) model rat with heart-kidney yang deficiency, and the sa me time approaching the mechanism of brady Arrhythmia model rat with heart-kidney y ang deficiencyMethods:In this study,we used heart-kidney yang deficiency molding method a ttributed to intraperitoneal injection of adriamycin and Hydrocortisone to Brady arrhythmia (Ba) rat model with heart-kidney yang deficiency. According to the maximum dose of xixin and fuzi recorded in Chinese Pharmacopoeia2010,we applied increasing-decreasing design method with baseline geometric proportion to regulate dose proportion of xixin and fuzi we got five different proportions of xixin and fuzi including groups Ⅰ (0:18)Ⅱ(6:12),Ⅲ(9. According to the ma ximum dose of xixin and fuzi recorded in Chinese Pharmacopoeia2010,we app lied increasing-decreasing design method with baseline geometric proportion to regulate dose proportion of xixin and fuzi.we got five different proportions of xixin and fuzi including groups Ⅰ (0:18),Ⅱ(6:12),Ⅲ(9:9),IV(12:6),and Ⅴ(18:0).The brady Arrhythmia model rats with heart-kidney yang deficiency were treated with five different proportions of xixin and fuzi.①Observing pathological morp hology changes of heart-kidney tissue by visual inspection②Record groups of electrocardiogram (ecg) in rats③ELISA was used to detect the serum content of Na+-K+-ATPase、MDA、SOD、and cTnI.Result:Compared with blank control group,Ba model group rats had significan t difference in Heart rate(P<0.05),serum content of MDA、cTnI and cGMP incr eased significantly(P<0.05),serum content of SOD、Na+-K+-ATPase and cAMP d ecreased significantly(P<0.05).Compared with Ba model group:①Group Ⅲand Ⅳhad obvious effect of significant difference in heart rate(P<0.05);②All the di fferent proportions groups could down-regulate MDA and cTnI and up-regulate the Antioxidant capacity factor SOD;groups of Ⅲ(9:9)andIV(9:12)had obvious e ffect of down-regulating the expression of MDA and cTnI (P<0.05);groups of Ⅲ(9:9)andIV(9:12) had obvious effect of up-regulating the expression of SOD (P<0.05);③All the different proportions groups could up-regulate Na+-K+-ATPas-e;groups of Ⅲ(9:9)andIV(9:12)had obvious effect of up-regulating the expressio-n of Na+-K+-ATPase (P<0.05)④All the different proportions groups could up-r-egulate cGMP,groups ofⅢ(9:9)andIV(9:12) had obvious effect of up-regulating the expression of cGMP(P<0.05);all the different proportions groups could do wn-regulate cAMP,groups of111(9:9) had obvious effect of down-regulating the expression of cAMP(P<0.05).Conclusion:①It was successful to replicate Ba rats model with heart-kidney y-ang deficiency molding method attributed toI ntraperitoneal injection of adriam-ycin and Hydrocortisone to Brady arrhythmia (Ba) rat model with heart-kidney-yang deficiency estimating from molding factors,common manifestation,objectiv e indicator,Counterevidence therapy and stability of model.②Different proportio ns of xixin and fuzi had certain regulating effect on Antioxidant capacity(SOD, MDA,cTnI) and Adjust the neuroendocrine function(Na+-K+-ATPase)of Brady a rrhythmia (Ba) model rats with heart-kidney yang deficiency.By down-regulatin g the expression of MDA,cTnI,cGMP and up-regulating the expression of SOD, Na+-K+-ATPase,cAMP,and then Enhance myocardial tissue anti-oxidation abilit y, regulating neuroendocrine functionmay be its therapeutic mechanism.③Grou ps of Ⅲ,Ⅳhad certain regulating effect on Enhance myocardial tissue anti-oxid ation ability, regulating neuroendocrine function Brady arrhythmia (Ba) model r ats with heart-kidney yang deficiency,but the groupⅣ had more comprehensive effect which may be related to its efficacy of restore Yang and salvage the bre akdown.The results showed that there were different therapeutical effect of diffe rent proportions of xixin and fuzi on Brady arrhythmia (Ba) model rats with h eart-kidney yang deficiency.Dosage proportions of xixin and fuzi have certain d ose-effect relationship with curative effect.This study provided experiment basis for clinic treatment of xixin and fuzi on Ba.
