Blood lipids in the serum is too high,it will lead to increased viscosity,if the blood vessels accumulate a large number of lipids will form plaque,and thus blocking blood vessels cause atherosclerosis.Atherosclerosis will further slow the blood flow of the coronary arteries,narrowing the blood vessels,reducing the amount of myocardial perfusion,resulting in myocardial ischemia or forming coronary heart disease.At present,the clinical use of lipid-lowering drugs for chemical synthesis,although it can effectively reduce blood lipids,but will produce certain toxic side effects,the safety of people to ponder.Therefore,the research and development of low toxicity,safe lipid-lowering health care products and functional food is of great significance.The soluble dietary fiber and pigment are rich in jujube peel,and they can clear free radicals and has good inhibition ability to lipid peroxidation,and it shows good antioxidant activity,but the effect of red date pigment and soluble dietary fiber of jujube on lowering blood lipid is not reported.In this experiment,jujube pigment,jujube soluble dietary fiber as raw material,to study the synergistic effect of red jujube pigment and soluble dietary fiber of red jujube on lipid peroxidation,to study the synergistic adsorption of cholesterol,cholic acid sodium and the activity of inhibiting pancreatic lipase and cholesterol esterase with the soluble dietary fiber of jujube and jujube,and to induce hyperlipidemia mice by high fat diet.To study the effect of reducing blood lipid and its synergistic effect on the soluble dietary fiber of red jujube and JuJube,and to provide some theoretical basis for developing natural antioxidants and functional health foods.The experimental results are as follows:(1)To study the synergistic effect of red jujube pigment and soluble dietary fiber of red jujube on lipid peroxidation.The results showed that the soluble dietary fiber of red jujube and JuJube was inhibited by lipid peroxidation,the inhibitory ability of soluble dietary fiber on lipid peroxidation of linoleic acid was stronger than that of red jujube pigment,and the soluble dietary fiber of jujube and jujube could inhibit the spontaneous lipid peroxidation of liver tissue in mice.,the inhibition ability of red jujube pigment is stronger than soluble dietary fiber and complex,the IC50 of pigment is 4.383 mg/ml,and the IC50 of soluble dietary fiber is 10.150 mg/ml.The ability of H2O2-induced spontaneous lipid peroxidation in mice was inhibited by the combination of red jujube pigment and soluble dietary fiber.Moreover,the inhibitory effect of red jujube pigment on H2O2 induced liver tissue lipid peroxidation and the inhibition ability of complex ligand in low concentration,and with the increase of concentration,the inhibition ability of red jujube pigment was stronger than that of jujube soluble dietary fiber and complex;the effect of red jujube pigment on Fe2+H2O2-induced liver tissue lipid peroxidation was obvious,and the inhibitory ability of soluble dietary fiber was weaker with the increase of concentration,and the inhibition was stronger than that of red jujube pigment.The red jujube pigment and the soluble dietary fiber of jujube were protected by H2O2-induced erythrocyte membrane lipid peroxidation and erythrocyte membrane hemolysis,and the soluble dietary fiber of red jujube and red JuJube had a significant inhibitory effect on the lipid peroxidation of erythrocyte membrane induced by H2O2.Using the principle of middle effect,when the inhibition rate within a certain range,the combined index is less than 1,jujube pigment and jujube soluble dietary fiber has synergistic effect,the share index is equal to 1,the two have additive effect,the combined index is greater than 1,the two do not have synergistic effect,Therefore,the soluble dietary fiber of jujube pigment and JuJube has the ability of synergistic lipid peroxidation under certain concentration.(2)To study the synergistic effect of red jujube pigment and soluble dietary fiber of jujube on blood lipid in vitro.The results showed that the soluble dietary fiber of red jujube and red JuJube had the ability of absorbing fatty acid,and the adsorption ability of red jujube pigment was stronger than that of jujube soluble dietary fiber,and the adsorption ability of the two dates was enhanced obviously.The combination of red jujube pigment and jujube soluble dietary fiber Cholic acid sodium has certain adsorption ability,and the adsorption capacity is as follows:Mixture>Red Date pigment>Date soluble dietary fiber;synergistic effect of red jujube pigment and soluble dietary fiber of jujube on the activity of pancreatic lipase and cholesterol esterase was significantly inhibited,and the inhibition ability of red jujube pigment was stronger than that of jujube soluble dietary fiber,using the principle of middle effect,When the inhibition rate is greater than 0.4,the combined index is less than 1,and the red jujube pigment and the SDF have some synergistic inhibition,and with the enhancement of action effect,synergistic effect enhancement,in vitro simulation of human intestinal environment,jujube pigment and jujube soluble dietary fiber alone and combined can reduce the solubility of cholesterol in the solution micelle,with the increase of concentration,the solubility of the alcohols in micellar was reduced,and the dose dependence was present.The results showed that the combination of red jujube pigment and soluble dietary fiber of red jujube had a certain effect on reducing blood lipid in vitro.(3)To study the synergistic effect of red jujube pigment and soluble dietary fiber of jujube on lowering blood lipid in vivo.The results showed that the total cholesterol content in the serum of mice was significantly elevated to 7.90±0.08 mmol/1(p<0.01).Compared with the blank group,the total cholesterol,triglyceride,LDL cholesterol in the serum of the hyperlipidemia group,The content of apolipoprotein A was significantly elevated,and the content of high-density lipoprotein cholesterol and apolipoprotein B was significantly decreased(p<0.01).Compared with the high fat Group,low dosage Group of red jujube pigment and soluble dietary fiber of jujube had no significant effect on lipid level and lipid level in hyperlipidemia mice(p>0.05),and the high dosage group of red jujube pigment and soluble dietary fiber of red jujube could significantly reduce total cholesterol and triglyceride in serum.Low density lipoprotein cholesterol,apolipoprotein a content,increased HDL cholesterol,apolipoprotein B content,and can reduce the mouse atherosclerosis index,the formation of atherosclerosis has a certain preventive effect and high dosage group of red jujube pigment and high dose of soluble dietary fiber of red jujube,total cholesterol in liver tissue of hyperlipidemia mice,triglyceride,high-density lipoprotein cholesterol,LDL cholesterol content also has a certain effect,and can improve the fat mice liver lipoprotein lipase,liver esterase and total lipase activity,It is more effective to use the two together.The results showed that the soluble dietary fiber of red jujube and red JuJube had the function of lowering blood lipid,and the joint use had synergistic effect.
