Polymorphism Of Apolipoprotein E Gene And Warfarin Dosage Effect In Han And Uygur Populations With Atrial Fibrillation In Xinjiang

Objective: To study the association between apolipoprotein E(APOE)gene polymorphism and oral warfarin effective dose in atrial fibrillation patients in Han and Uygur populations of Xinjiang Region.Methods: The first affiliated hospital of Xinjiang Medical University,100 cases of Uygur and Han patients with atrial fibrillation were selected as the case group;100 cases of Han and Uygur non-AF were used as control group in the health management center at the same time period.All of 400 cases of venous blood DNA,PCR-RFLP method was used to detect the gene polymorphism of rs429358 in APOE gene.The genotypes and allele frequencies were calculated and compared between two ethnic groups APOE gene polymorphism and the maintenance dose of warfarin orally administered to two ethnic AF patients in two ethnic groups in China.Results: The frequencies of genotypes E2 / E2,E2 / E3,E2 / E4,E3 / E3,E3 / E4 and E4 / E4 of APOE gene rs429358 in Xinjiang were 0.75%,8.75% %,77.5%,10% and 1.5%,respectively.The proportions of E2,E3 and E4 alleles were 9.5%,79% and11.5% respectively.There was no significant difference in the maintenance dose of warfarin among different ethnic groups(P> 0.05).Warfarin with high E2 / E3 genotype maintenance dose and warfarin E3/E4 genotype low maintenance dose.The effective dose of warfarin was significantly different among E2/E3,E3/E4 andE3/E3 genotypes in patients with Han nationality atrial fibrillation(P<0.05).Compared with the three genotypes of patients with Uygur atrial fibrillation,warfarin effective dose was significantly different(P value <0.05).Conclusion: The rs429358 polymorphism of APOE gene in Xinjiang may be one of the influencing factors of warfarin dose in AF patients.
