Objective:This work was set to illustrate the absorption,metabolism,distribution and elimination of dihydromyricetin and myricetin from Hovenia acerba Lidl.in rats.Methods:After Hovenia acerba lindl.extract was administered orally to rats,the plasma,bile,urinary,fecal and organ samples in rats were collected at different time points.All samples were analyzed by UHPLC-MS/MS after pretreatment.Results:Dihydromyricetin and myricetin in the Hovenia acerba Lindl.were dete-cted in the systemic circulation,and the metabolites of I and II phases were not iden-tified in the plasma samples.It was not directly deduced whether the metabolic trans-formation of dihydromyricetin and myricetin occurred in the blood.Dihydromyricet-in in plasma was detected at 0.5-2 h,which can be inferred that its absorption and eliminaion were fast.Myricetin in plasma was detected at 0.5-4 h,which indicated the absorption was fast.In the liver,dihydromyricetin and its I-phase metabolites and myricetin were detected with higher abundance,dihydromyricetin and its metabolites of phase I and ? as well as myricetin were detected in bile,it showed that dihydrom-yricetin and myricetin were distributed in the liver,and dihydromyricetin underwent biotransformation in the liver.The undetectable dihydromyricetin and myricetin in brain samples presented that they can not penetrate the blood-brain barrier.Dihydro-myricetin and myricetin were detected in the heart,and their metabolites were not detected,and it was not possible to deduce whether the metabolism of dihydromyri-cetin or myricetin in the blood taken place in the heart.In the kidneys,dihydromyri-cetin and its phase I metabolites and myricetin were detected.The presence of dihyd-romyricetin and its metabolites of phase I and myricetin was found in urine and feces of rats,with a higher abundance of dihydromyricetin in the feces.Simultaneously myricetin intestinal metabolites were detected in the feces.Conclusion:Dihydromyricetin and myricetin were detected in the plasma,bile,heart,liver,kidney,urinary and feces samples of rats.Phase I metabolites of DMY were detected in the liver,kidney and urinary samples of rats.Phase I,II metabolites of DMY were detected in bile of rats.Intestinal metabolits of MY were detected in feces of rats.Distribution of dihydromyricetin and myricetin and their metabolites in blood,bile,and organ samples of rats after oral administration of the extract ofHove-nia acerba Lindl.using UHPLC-MS/MS can be inferred that dihydromyricetin bio-transformed in the liver and kidney,and excreted through the urine and feces.Myrice-tin was not biotransformed in the liver and also excreted through the urine and feces.It showed that dihydromyricetin and myricetin were distributed in the liver,and the function of sober up and liver protection does have its material basis,dihydromyrice-tin and myricetin quickly absorped,widely distributed,dihydromyricetin biotransfor-mation occurred in the liver and kidney.
