Object:1.To explore the effect of the prescription of Juanbilijieqing on inflammatory factor IL-8 in the synovial membrane of iliac joint in rats with gouty arthritis.2.To increase the expression of MMP-9 in macrophages based on MSU crystals.Qingfang Regulates the Effect of MMP-9in Synovial Tissue of Rats with Gouty Arthritis.Methods:A rat model and 72 SPF rats were used.The gouty arthritis model was prepared by continuous intraperitoneal injection of potassium oxonate solution and modified Coderre method for 7 days.The rats were randomly divided into low-dose groups(group A).Medium-dose group(B group),high-dose group(C group),western medicine foxifene group(D group),model group(E group),blank group(F group),observed in each group after modeling Swelling around the ankle,blood uric acid,synovial tissue IL-8,MMP-9 changes.Results:After the modeling,the rats in the low,medium and high doses of the Chinese medicine group and the model group showed marked swelling in the sacroiliac joint,and the swelling index,IL-8 and MMP-9 expression of the joints were significantly decreased(P<0.05).The serum uric acid value was significantly lower,and the high-dose group decreased more significantly.Conclusions:1.The prescription of Juanbilijieqing can reduce the expression of IL-8,but the specific mechanism is through inhibiting Toll/Myd88/NF-?B pathway or the formation of NALP3/caspase-1/ACS complex.Lowering the expression of IL-8 still requires further study.2.The mechanism by which the prescription of Juanbilijieqing reduces MMP-9 by inhibiting the inflammatory chemotaxis of IL-8 and inhibiting the secretion of 92kDa zymogen from inflammatory infiltrated PMN(polymorphonuclear leukocyte)and reducing pro-MMP The activation of-9 affects the expression of MMP-9.Still,by inhibiting the synthesis of EK1/2 kinase in macrophages and inhibiting the activation of precursor pro-MMP-9.further exploration is needed to provide a new direction for the treatment of gout in the future.
