Study On The Relationship Between TCM Constitution Theory And Gene Expression Of Osteoporosis

Objective : This study investigated the general situation of Osteoporosis Epidemiology and TCM Constitution,determine easily induced OP TCM type of constitution.To investigate the osteoporosis of different types of TCM Constitution respectively with vitamin D receptor(VDR)gene(Bsm locus I)polymorphism,type I collagen(COLIa I)gene(Bal I locus)the relationship between the polymorphism and bone mineral density,type and different constitution,relationship between gene polymorphism of OP,not only for screening the susceptible population of TCM and provide a theoretical basis for diagnosis and treatment,different OP patients to provide more clinical objective standard.Methods : From February 2015 to December 2016 were collected from the Department of orthopedics of Shanxi People's Hospital and included patients with osteoporosis consistent standard,aged 55 to 85 years old,the average(71.83 + 6.89)years of age,physical differentiation,choose Yang deficiency in Yin deficiency group,blood stasis group,and group the 4 constitution group were 179 cases patients as experimental group,were studied,analyzed.Yang deficiency group had 95 cases,the mean age of(71.08 + 6.98)years old.41 cases of yin deficiency group,the average(69.07 + 7.18)years old.Blood stasis group 24 cases,the average(67.83 + 6.08).The gentleness group 19 cases,the average(69.16 + 7.11)years old.At the same time collection of unrelated healthy volunteers,blood samples,and syndrome differentiation of volunteers,selected from the group of Yang deficiency,yin deficiency group,blood stasis group,and group the 4 constitution group were 54 cases as control group,aged 55 to 85 years old,the average(67.20 + 7.21)years old.Yang deficiency group 19 cases,16 cases of yin deficiency group,blood stasis group 6 cases,the gentleness group 13 cases.The Shanxi People's Hospital using dual energy X-ray absorptiometry,respectively in patients with lumbar and femoral neck BMD the value of detection and analysis All of the patients.In the experimental group,through the use of PCR-RFLP technology to study the vitamin D receptor,gene polymorphism,type I collagen analysis.Excel table is used to record the data into the table,through the analysis of SPSS19.0 software,with the result of the analysis to determine the incidence of health.Methods the patients and their families to participate in inform the specific test,and signed informed consent.Results:(1)OP patients,the largest number of Yang deficiency group,significantly more than the other group of the constitution;(2)the bone density and body mass,relationship between gene polymorphism: Yang deficiency groups had lower BMD values generally,bone density in different groups of fitness value comparison,there were statistically significant difference(P < 0.05);VDR gene BB,Bb,bb three kinds of genotype L2-4,bone mineral density Neck value: BB genotype L2-4,Neck,BMD value is lower than the Bb,BB BMD,three genotypes of BMD is no significant difference(P > 0.05);Type I collagen detected only SS genotype was not detected polymorphism COLIaI gene in Sp1 loci;(3)the distribution of VDR genotype in OP patients: the numberConclusion:(1)OP in the patients with Yang deficiency group and yin deficiency group,the largest,compared to other physical group,Yang deficiency and yin deficiency in the population with OP would be more likely;(2)polymorphism of VDR gene,the type of TCM constitution is different,can affect the BMD value of important the factors change;(3)COLIa I gene detected only SS genotype,so it can not explain the polypeptide gene is able to change the value of the influence factors of different constitution types in patients with BMD;(4)although the different types of TCM Constitution can be based on the "Chinese constitution classification and determination of self Testing > dialectical,but the subjective judgment of human still accounted for most,making the diagnosis of the disease is not objective.This thesis research the relationship of gene and constitution,and provide a theoretical basis for clinical more objective and more accurate constitution.
