Objective:To observe the effect of Pingchuan Granule on the level of EOS in BALF and the expression of miRNA-21 in chronic asthmatic rats,and to explore the mechanism of the effect of Pingchuan Granule on the EOS level in BALF of chronic asthmatic rats by regulating miRNA-21 expression.Methods:1.80 Wistar rats were randomly divided into normal group,model group,dexamethasone group and Chinese medicine group.The rats in the normal group were given normal saline atomization,and the rest groups established chronic asthmatic rat models with ovalbumin.The traditional Chinese medicine group was given Pingchuan Granule(0.45mg/Kg)for gastric perfusion,and dexamethasone was given the liquid of dexamethasone in the group of dexamethasone.The normal group and the model group were given the liquid of traditional Chinese medicine.To observe the effect of Pingchuan Granule on lung tissue and EOS infiltration in chronic asthmatic rats.Chronic inflammation and infiltration of EOS in chronic asthmatic rats were examined under light microscope.Realtime-PCR was used to detect the expression level of miRNA-21 in lung tissue.The EOS counts in the rat BALF were detected under the microscope.To explore the effect of Pingchuan Granule on EOS level in BALF of chronic asthmatic rats.2.100 Wistar rats were randomly divided into normal group(group A),model group(group B),miRNA-21 inhibition expression group(C group),miRNA-21 inhibition expression vector control group(D group)and traditional Chinese medicine group(E group).Group A was given normal saline atomization,and B,C,D and E groups caused chronic asthma animal models.Group C and D were injected with corresponding lentivirus solution by tail vein before modeling,0.1ml each time.Once every three days,four times.Group E received Pingchuan Granule,and the method and procedure were same as experiment 1.The EOS counts in the rat BALF were detected under the microscope.To investigate the effect of Pingchuan Granule on the level of EOS in BALF of chronic asthmatic rats by inhibiting the expression of miRNA-21.Results:1.Pingchuan Granule can reduce inflammatory infiltration in lung tissue of chronic asthmatic rats and improve the pathological state of lung.2.Pingchuan Granule can reduce the EOS count in BALF of chronic asthmatic rats and reduce the degree of EOS infiltration.The average EOS percentage of lung tissue in each field was 8.40 ± 1.60%.Compared with the model group,there was a significant difference(P<0.05),but there was no significant difference compared with the dexamethasone group(P>0.05).3.Pingchuan Granule could reduce the expression of miRNA-21 in lung tissue of chronic asthmatic rats,and there was a significant difference compared with that of the model group(P<0.05),and there was no significant difference compared with that of dexamethasone group(P>0.05).4.Pingchuan Granule can reduce the EOS count in BALF and decrease the degree of EOS infiltration in chronic asthmatic rats by inhibiting the expression of miRNA-21.The average percentage of EOS infiltration in each lung tissue was 7.85 ± 0.81%,compared with the normal group(P<0.05),and there was significant difference(P<0.05)compared with the model group(P<0.05).There was a significant difference compared with the miRNA-21 inhibition expression vector control group(P<0.05),and there was no significant difference compared with the miRNA-21 inhibition expression group(P>0.05).Conclusions:1.Pingchuan Granule can alleviate chronic inflammation and infiltration of EOS in chronic asthmatic rats.2.Pingchuan Granule can inhibit the expression of miRNA-21 in chronic asthmatic rats.3.Pingchuan Granule can reduce the EOS count in BALF of chronic asthmatic rats.4.Pingchuan Granule regulates the level of EOS in BALF of chronic asthmatic rats by inhibiting the expression of miRNA-21.
