Expression Of MiRNA-21 And Eosinophil In Patients With Chronic Rhinosinusitis And Its Clinical Study

Objective:To compare the levels of miRNA-21 and eosinophils in patients with chronic rhinosinusitis and the normal population,and to explore the role and related mechanisms of miRNA-21 in the occurrence and development of chronic rhinosinusitis.The diagnosis and treatment of chronic rhino-sinusitis provide new research ideas;on the other hand,by analyzing the correlation between miRNA-21 and eosinophils,the source of miRNA-21in serum is explored,which is eosinophils and miRNAs-21's relationship provides evidence.Methods:According to the inclusion criteria,exclusion criteria,and classification of chronic rhinosinusitis,a total of 30 eligible patients with chronic rhinosinusitis(including 13 cases of CRSsNP type and 17 cases of CRSwNP type)were collected as the experimental group and the nasal septum was simply diagnosed Twenty patients with deflection were the control group.Real-time fluorescent quantitative PCR was used to detect the levels of miRNA-21 in serum and the levels of eosinophils in whole blood were measured for comparison.Results:1.The average level of serum miRNA-21 in the CRSwNP group was 1.54±0.16,the average level of serum miRNA-21 in the CRSsNP group was 1.10±0.19,and the average level of serum miRNA-21 in the normal control group was 1.04±0.20.It was found that there was a statistically significant difference in serum miRNA-21 levels between the CRSwNP group and the normal control group(P<0.001).2.3(23.08%)patients with CRSsNP were positive for eosinophils in peripheral blood,9(52.94%)patients with CRSwNP were positive for eosinophils in peripheral blood,and 1in the control group(5.0%)were positive,and the differences between the three groups were statistically significant(?2=10.828,P=0.004).Pairwise comparison found that the difference between the CRSwNP group and the control group was statistically significant(P<0.05).3.Compare the diagnostic value of ROC curve of miRNA-21 and eosinophils in the CRS group with the control group,the areas under the curve of miRNA-21 and eosinophils are 0.482,0.829,the area under the curve of miRNA-21 is<0.5,and the area under the eosinophil curve is between 0.7-0.9,which has a certain accuracy.At the maximum Jordanian index,the corresponding eosinophil concentration cutoff was 0.25×10~9/L,its sensitivity was 76.5%,and its specificity was 78.8%.4.The serum miRNA-21 and eosinophils in the CRS group were analyzed by Spearman rank correlation,the levels of serum miRNA-21 and eosinophils in the CRS group were statistically significant(r=0.406,P<0.05).Conclusion:1.miRNA-21 is related to the occurrence and development of CRS,especially CRSwNP,indicating that serum miRNA-21 may be involved in the pathogenesis of CRS.2.Eosinophils are involved in the pathogenesis of CRS and can be used as a reference indicator for the diagnosis of CRS.3.Eosinophils are of great value in the diagnosis of diseases,while miRNA-21 is of little value in the diagnosis of CRS.Eosinophils may be used as auxiliary biological indicators for the diagnosis of CRS diseases.4.There is a overall trend between serum miRNA-21 and eosinophils levels,which provides evidence for the source of miRNA-21 and provides research directions for the mechanism of miRNA-21 participating in CRS.
