Purpose:By establishing model of knee osteoarthritis,to observe the effect of Aconitum injection on the contents of TNF-?,COX-2 and PGE2 in the synovial fluid of knee osteoarthritis model rabbits,and study the preliminary mechanism of Aconitum injection in the treatment of osteoarthritis.Method:40 New Zealand white rabbits were randomly divided into blank group,model group,sodium hyalurate group and aconitum injection group.In addition to the blank group,the other three groups were injected papain into the knee joint to establish the knee osteoarthritis mo del.After the model was successfully manufactured,according to 0.1 ml/kg,sodium hyaluronate was injected into the knee joint of rabbits in sodium hyaluronate group,and aconitum injection was injected into the knee joint in the aconitum injection group,and the normal saline was injected into the knee joint cavity of the model group on day 1,day 4,day 7 and day 10.After another week,four groups of white rabbits were sacrificed by air embolism,and the articular fluid of each group was extracted and the content of TNF-?,COX-2 and PGE2 in the articular fluid was determined by Elisa method;and their articular cartilage tissue was removed.Then the his to morphologic changes of articular cartilage of rabbit knee were observed under microscope and scored according to Mankin scale.Result:The total score of the Mankin score of the joint cartilage of aconitum injection group was compared with that of sodium hyaluronate group and model group P<0.05,and the differences was statistically significant.The content of TNF-?,COX-2 and PGE2 in the joint fluid of aconitum injection group was compared with that of the model group and sodium hyaluronate group P<0.05,and the differences was statistically significant.Conclusion:Aconitum injection can improve the destruction of articular cartilage in rabbit knee osteoarthritis model,and has a protective effect on articular cartilage,which may be related to the inhibition of TNF-?,COX-2andPGE2 expression in articular fluid.
