Objective: The content of AXIN and TGF-?1 in articular cartilage of knee osteoarthritis and their protein expression were detected by experimental study.The therapeutic effect of aconite injection on KOA was studied at gene level,which provided theoretical basis for clinical application.Method: 1.34 male New Zealand big ear rabbits were randomly divided into blank group and model group.There were 9 rabbits in blank group and 25 rabbits in model group.In the model group,2% w/v papain and 0.03mol/L-cysteine(2:1)were injected into the knee joints on the first,third and fifth days respectively.The dosage was calculated as 0.1ml/kg.After 2 weeks of normal feeding,a rabbit was randomly selected from the blank group and the model group for dissection.The degeneration of articular cartilage was observed with naked eyes,and pathological sections were observed under the microscope to confirm t he success of the model.Then 24 rabbits in the model group were randomly divided into model group,control group and experimental group,8 rabbits in each group.2.After the successful establishment of the model,on the 1st,4th,7th and 10 th days,the blank group was not disposed of,the model group was given saline,the control group was given sodium hyaluronate injection,and the experimental group was given Aconitum injection?3.The rabbits was killed by air embolization,and the changes of cartilage were observed by HE staining;the mRNA expression of AXIN and TGF-? 1 was detected by PCR;and the protein expression of AXIN and TGF-? 1 was detected by immunohistochemistry.4.SPSS19.0 was used to analyze the data.The measurement data was expressed as(x±s).The comparison between groups was analyzed by one-way anova.P < 0.05 indicated that the results had statistical significance.Result:1.Naked eye observation: compared with other groups,the destruction of articular cartilage in the experimental group was mild,most of them were white,the surface of cartilage was glossy,and there were few cracks and defects.2.HE staining under light microscopy: The cytoplasm of chondrocytes in the experimental group was pink and the nucleus was blue.Compared with other groups,the structure of soft tissue tended to be normal,but the arrangement was disordered but the layers were separable.The number of chondrocytes increased locally,and the tide line was obscure.3.Immunohistochemical analysis: The expression of AXIN and TGF-?1 in the model group was significantly higher than that in the blank group(P < 0.05).The expression of AXIN and TGF-?1 in the control group was significantly lower than that in the model group(P < 0.05).The expression of AXIN and TGF-?1 in the experimental group was significantly lower than that in the model group(P < 0.05).Learning Significance.The expression of AXIN and TGF-?1 in the experimental group was similar to that in the control group(P > 0.05),but there was no significant difference between the two groups.4.PCR analysis: The AXIN and TGF-?1 in the model group were significantly higher than those in the blank group(P < 0.05),the difference was statistically significant;the AXIN and TGF-?1 in the control group were significantly lower than those in the blank group(P < 0.05),the difference was statistically significant;the AXIN and TGF-?1 in the experimental group were significantly lower than those in the blank group(P < 0.05),and the difference was statistically significant.The levels of AXIN and TGF-?1 in the experimental group were significantly higher than those in the control group(P < 0.05).Conclusion:Aconitum injection can reduce the protein ?m RNA expression of AXIN and TGF-?1 in KOA articular cartilage,inhibit the degradation of articular cartilage and protect articular cartilage.
