Objective:Through the experimental study of this subject,To investigate the effect of Aconitum injection on IL-1? and IL-18 in synovial fluid of knee osteoarthritis in rabbits,Lay the foundation for further clinical application.Method:1?A mixed solution of 2% w/v papain solution and 0.03 mol/L L-cysteine solution ratio of 2:1,Press 0.1ml/kg into New Zealand White Rabbit Double Posterior Knee Joint Cavity,Establishment of rabbit knee osteoarthritis model.2?Twenty male New Zealand white rabbits were randomly divided into body weight numbers:Blank group,Model group,Positive drug control group,Experimental group.Except blank group,other groups to model,2 weeks after the X-ray observation of the success of modeling.After modeling success,Start treatment.The experimental group was injected with aconitine injection 0.2ml,Positive drug control group injection of sodium hyaluronate injection 0.2ml,The model group was injected with 0.2ml of saline,Blank group does not give any treatment.After 1 week of treatment,take the posterior knee joint fluid,The levels of IL-1? and IL-18 in synovial fluid were measured;Anatomy of the knee;Observed by HE staining microscope.3?Statistical analysis software SPSS19.0 to deal with the data,If P<0.05 that the test results were statistically significant.Count data with the composition ratio or rate of the form that need to use x2 test;measurement data with x ąs form that need to use t test?Results:1?Visual observation of cartilage results:Compared with the blank group,the model group found that there were many cracks or defects on the cartilage surface,The edge of the joint almost all the formation of osteophytes,and was yellow;Positive drug control group and experimental group were similar,Compared with the model group,the cartilage surface crack or defect was found to be less,Little osteophyte formation,and was white.2?Microscopic observation Mankin score results:The scores of the model group were higher than those of the blank group(P<0.05),the difference was statistically significant.The positive control group and the experimental group were lower than the model group(P<0.05),The difference was statistically significant.3?The levels of IL-1? and IL-18 in synovial fluid of model group were significantly higher than those in blank group(P<0.05),The difference was statistically significant.Indicating that the experiment was successful;The levels of IL-1? and IL-18 in the synovial fluid of the control group and the experimental group were significantly lower than those in the model group(P<0.05),The difference was statistically significant.Conclusion:Aconite injection not only play the role of protection of cartilage and can reduce the knee joint osteoarthritis synovial fluid IL-1? and IL-18 content,To achieve the purpose of eliminating inflammation.
