Objective: To establish a metabolic syndrome model in Sprague Dawley(SD)rats.To investigate the changes of prostatic tissue and expression level of 5?-reductasetype ?(SRD5A2)in SD rats receiving high-fat diet and Insulin resistance agent(Metformin)intervention.Methods: We have established metabolic syndrome(Met S)models by providing rats with high fat diet,to intervention with Metform in was also applied.All objects were sacrificed 40 days later with prostatic tissue being removed,weighed before stained,fasting blood glucose(FBG),fasting blood insulin(FINS),homeostasis model assessment of insulin resistance(HOMA-IR),body weight,prostate tissue weight and prostate index were obtained.Prostate tissue were kept for histology.The expression of SRD5A2 was determined by western-blot and immunocy to chemistry.Results: Metabolic syndrome was induced by high-fat diet successfully,significant increase in FINS and HOMA-IR(p<0.05)were found in high-fat diet than normal diet,and prostatic hyperplasia occurred in this model,significant increase in weight,prostate weight and prostate index(p<0.05)were found in it.However,with metformin intervene,significant decrease was found on FINS,HOMA-IR,body weight,prostate weight and prostate index(p<0.05)compared with that of the high-fat diet.The expression of SRD5A2 in hyper plastic prostate tissue was increased,and was positively associated with the degree of hyperplasia of prostate.Conclusions:The successful Met S model caused by high-fat diet was validated by Insulin resistance,prostatic hyperplasia occurred in those model animals.Metform in agents can be used to treat BPH caused by metabolic syndrome.The expression levels of SRD5A2 have positive correlation with the level of prostatic hyperplasia.Met S might be a potential pathogenesis of benign prostatic hyperplasia through the SRD5A2 pathway.
