Oplopanax elatus Nakai is one of the rare medicinal plants in Changbai Mountain,which has various medicinal values.In recent years,due to the serious damage to its wild resources,and the immature artificial cultivation system,there is a serious shortage of plant materials in O.elatus,and the development of related products is limited.Therefore,it is urgent to seek an efficient way to obtain a large amount of plant materials quickly.Plant tissue culture is the core of the endangered medicinal plants save and propagation techniques.Using this method to quickly obtain a large number of O.elatus adventitious roots as a new type of plant material,which will have important social significance.Although there are antioxidant and anti-inflammatory properties in O.elatus,the research is still in its infancy.Therefore,extracts were obtained from adventitious roots of O.elatus cultured in bioreactors was as the research material of this study.Extracting adventitious roots through different polar organic solvents to prepare extracts to investigate anti-inflammatory andantioxidant properties.And it will provide a theoretical basis for the development of related products from the O.elatus.In the antioxidant experiment,the DPPH scavenging ability,chelating ability,and reducing power were measured to evaluate the antioxidant properties of O.elatus extracts.All extracts prepared by methanol(CE),n-hexane(n-Hex),dichloromethane(DCM),ethyl acetate(EtOAc),n-butyl alcohol(n-BuOH),and water layer condensate(Aqueous)showed antioxidant properties with a dose-dependent manner.Among the.extracts,the minimum IC50 value of DPPH scavenging ability or chelating ability,and the maximum OD value of reducing power were found in EtOAc,which indicated that the ethyl acetate extract possessed a high antioxidant capacity.The anti-inflammatory effect of CE on LPS-induced ALI model was conducted.The CE of 200 mg kg-1 significantly(p<0.01)inhibited the cell concentration and pro-inflammatory mediators including nitric oxide(NO)content,tumor necrosis factor-?(TNF-?),interleukin 6(IL-6)and interleukin 1?(IL-1?)in BALF.The histopathology of lung tissue was observed.The alveolar wall with less damage was found in the lung tissue treated by 200 mg kg-1 CE,and the inflammatory cell infiltration was also inhibited.CE increased the superoxide dismutase and glutathione peroxidase activities and reduced myeloperoxidase activity and malondialdehyde content.The present experiment indicated that methanolic extract of O.elatus adventitious roots had higher anti-inflamatory property.In the vitro anti-inflammatory experiment,the effect of six extracts(CE?n-Hex?DCM?EtOAc?n-BuOH and Aqueous)on NO content in LPS-induced peritoneal macrophage(PMs)was investigated.The NO content in Cell reduced in all treatemts of six extracts,the maximum inhibitory effect was determined in DCM and EtOAc.Therefore,the anti-inflammatory effect of DCM and EtOAc were futher investigated.Two extracts inhibited the formation of prostaglandin E2,TNF-a,IL-6 and IL-1? with a dose-dependent manner.The inhibition effect was also found in the expression of nitric oxide synthase and cyclooxygenase protein expression.In addition,the present study also investigated anti-inflammatory machanism of MAPK and NF-?B signaling pathways.In the MAPK signaling pathway,DCM and EtOAc inhibited protein phosphorylation of p3 8,JNK,and ERK.In the NF-?B signaling pathway,the expression of I?B-? was promoted by the DCM and EtOAc treatments.Meanwhile,the nuclear translocation of NF-?B was also inhibited.Findings of the present study revealed that DCM and EtOAc regulated the MAPK and NF-?B signaling pathway to act the anti-inflammatory effect.
