Oplopanax elatus Nakai is a perennial deciduous shrub of Araliaceae, narrowly distributed. There is only a small amont in China, Russia and North Korea all over the world. Its roots and stems are used as medicine for invigorating vital energy, strengthening yang and stimulating central nervous system. Clinically it is used to treat cardiovascular disease, diabetes, rheumatism, neurasthenia, mental depression, schizophrenia, impotence and so on. Oplopanax elatus Nakai is a kind of potential medical plant specially in the Northeast. However, in recent years, it has been in endangered status because of resource damadge. Therefore, studying the leaves profits us to make full use of this plant as well as protect it. This article has made full pharmaceutical research of total saponins extracted from Oplopanax elatus Nakai,including several parts as follows:1. Referring to the related literature, We reviewed the latest research progress of Oplopanax elatus Nakai in several aspects such as chemical constituents, pharmacological activity, clinical application and so on.2. We isolated one of the most saponins from Oplopanax elatus Nakai through silica gel and ODS column chromatography.Using it as reference substance, we could make qulitative and qutitative analysis of total saponins.3. During determination of the total saponins content of extracting material, we compared Vanillin- Perchloric acid chromogenic method with Vanillin- Concentrated sulfuric acid chromogenic method. In the former, the reference substance and the sample has the same maximum absorption wavelength, and besides, sensitivity of the former is higher, thus we chose it. After methodology validation, Vanillin- Perchloric acid chromogenic method is found applicable to determinate total saponins content and can be used to guide the research of extracting technology.4. We chose several factors that may influence results of the extraction, including concentration of ethanol, volume, time and times of extraction. First, we did single factor experiments to determinate the extent of these factors, and then orthogonal test showed that the best technology is reflux extration with 60% ethanol(8 folds) for 3 times ,half an hour once.Validation experiments were also done to make sure the technoly stable and applicable. 5. Macroporous adsorption resin were used to purify the total saponins. We referred to the literature and analyzed the structures of those saponins, then chose two kinds of resins, D101 and AB-8. They behaved almost the same on dynamic elution, but D101 has better static adsorption and desorption coeffiecient, Therefore, D101 was chosen.Then influence factors of purification such as amount and concentration of the sample, concentration and volume of the eluent,together with the eluting rate were evaluated. The result was as follows: The amount should be no more than 6g dried medicinal herb for per gram macroporous resin. 4 BV 20% ethanol is needed to get rid of aqueous impurities, after that, 6 BV 60% ethanol elutes the resin at the rate of 1 BV/h. Total saponins gained after drying, the content is more than 50%.6. Following Chinese Pharmacopoeia, we established quality standard of extrating material from three aspects as identification, inspection and determination. The identification was built with TLC method, the water and ash content were measured to set a limit, the content of total saponins was determinated and 80% of that is the limit.The next step was stability test, including influencing factor test,accelarating test and long-time test. Primary test shows extracting material stable, long-time test needs more time to get a result.7. Based on the pharmacology experiment and micromeritics properties,volume of the capsule can be decided. Meanwhile, we made quality control of Cirenshen Capsule according to Chinese pharmacopoeia, and the result is in line with the regulations.
