Objective:Through this experiment,to discuss the mechanism of Korean medicine prescription "Xiangfuzibawu decoction"'s action on liver depression and spleen deficiency syndrome,to provide animal experimental basis for clinical drug use,and to provide effective prescription of Korean medicine for treating liver depression and spleen deficiency syndrome.Methods:We selected the SD rats as the experimental object,adopted the methods of tail clamped stimulation,feeding disorder,DaHuang intragastric administration purgation,caused liver depression and spleen deficiency in rats,prepared a model of liver depression and spleen deficiency in experimental rats.Rats were randomly divided into four groups:normal control group,liver depression and spleen deficiency model group,positive control gourp(Xiaoyaosan group),Xiangfuzibawu decoction medication group,12rats in each group,male and female mixed.The general state and macroscopic syndrome of rats in each group were observed,and the amount of water intake,food intake and weight change of each group were recorded.After 16 days of model production,the rats of model group were filled with saline every day,the positive control group and the drug treatment group were given Xiaoyaosan?Xiangfuzibawu decoction enrichment decoct soup by intragastric gavage;After 14 days of gavage,each group of rats was given anaesthesia With the urethane by intramuscular injection.Then we Gathered the eyeball,blood sampling from the fundus vein,and then Drag the rats' neck to death at the last.We used ELISA detection method to determine the changes of the efficacy indexes of MTL,GAS and pepsin in the serum of rats.We dissect the abdomen and pick up the stomach,cut out a piece of the gastric mucosa with a severe injury,put in 10%formaldehyde solution and fix 24h.Then doing something about embedding,section,Haematoxylin staining and sealing piece.Under different magnification of microscope,we observed pathological morphological changes of rat' s gastric mucosa tissue,compared the groups to each other to explore the mechanism of action of Xiangfuzibawu decoction on liver depression and spleen deficiency syndrome.Result:The feeding amount of each group was decreased except the normal control group after the model was finished.Compared with the normal control group,the difference was statistically significant between the model group,the Xiaoyaosan group and the Xiangfuzibawu decoction group(P<0.05).After the treatment,there was a significant increase in the amount of food in both the Xiaoyaosan and Xiangfuzibawu decoction group,and the difference was statistically significant(P<0.05).Before building model,the quality of rats in normal control group,model group and drug treatment group were compared with each other,and the difference was not statistically significant.(P>0.05).After the completion of the manufacturing model,the body mass of the rats in the model group,the Xiaoyaosan group and the Xiangfuzibawu decoction group decreased significantly compared with the normal control group(P<0.05).After the drug treatment,the body mass of the Xiaoyaosan group and the Xiangfuzibawu decoction group was significantly increased,and the difference was statistically significant compared with the model group(P<0.05).There was no statistically significant difference between Xiaoyaosan and Xiangfuzibawu decoction group(P>0.05).The MTL,GAS and pepsin index in the rat's serum of the model group were significantly decreased compared with that of normal control group,with significant difference(P<0.05).The contents of MTL,GAS and pepsin in the serum of the rats of the Xiangfuzibawu decoction group and Xiaoyaosan group were significantly higher than those in the model group,with significant difference(P<0.05).The gastric mucosal epithelial cells of rats in the model group showed obvious shedding,the glandular arrangement was disorganized,the secretion of mucus increased,and there were some changes in interstitial edema,and the mucosal vascular dilation and hyperemia.The glandular arrangement of the Xiaoyaosan group was relatively loose compared with the normal control group,and the shedding of epithelial cells decreased significantly.The Xiangfuzibawu decoction group still had a small number of exfoliated cells and a small amount of mucus secretion,but the glandular arrangement was significantly more orderly than that of the model group.After drug treatment,the pathological changes of interstitial edema and hyperemia were significantly improved.Conclusion:Xiangfuzibawu decoction can significantly increase serum gastrin and motilin concentration in the serum of liver and spleen deficiency model,significantly increase serum pepsin content,and there is no significant difference compared with the positive control drug Xiaoyaosan.By regulating gastrointestinal hormones and pepsin,it can promote gastrointestinal peristalsis,promote digestion,and improve symptoms of liver depression and spleen deficiency.Xiangfuzibawu decoction can also improve the pathological changes in the lining of the intestines of the rat,which is caused by liver and spleen,and repair the damaged gastric mucosa,and improve the disorder of the gastrointestinal function.
