Purpose: To observe the effects of Ruanjian Xiaoying Decoction on thyroid hormone level,serum Th1/Th2 level and thyroid pathological morphology in experimental autoimmune thyroiditis(EAT)rats with liver depression and spleen deficiency syndrome,and to explore its mechanism,so as to provide deeper experimental basis and new ideas for the treatment of chronic lymphocytic thyroiditis.Material and method:Female SD rats of SPF grade were fed adaptively for 7 days.They were randomly divided into normal group and model group according to body weight level.Normal group animals were fed naturally without any stimulation.Immune-mediated combined with iodine-rich water induction was used to establish the disease model.On the8 th day,the rats in the normal group were given distilled water.On the 22 nd day,rats were immunized for the first time.Tg(Tg: CFA = 1:1)was fully emulsified by full Freund’s adjuvant(CFA)into oil-in-water Tg(Tg: CFA = 1:1)by subcutaneous injection at multiple points on both hind feet.Tg(Tg: CFA = 1:1)was injected twice,with an interval of two days,each containing TG100 On the 29th-56 th day,the rats were immunized with Tg emulsified by incomplete Freund’s adjuvant(IFA)and injected subcutaneously,once a week,with "chronic restraint stress + excessive fatigue + dietary disorder method".The rats were restrained in a restraint tube for 3 hours a day and were placed in a plastic bucket filled with warm water for20 minutes,swimming to the endurance limit in one day and prohibited every other day.Eat in full every other day.At the end of modeling,the rats were randomly divided into model group,Tripterygium wilfordii group,Ruanjian Xiaoying Decoction group and Jinkui Shenqi pill group.After 56 days of administration,rats were sacrificed and the following indicators were detected:(1)thyroid weight index(2)thyroid hormone level(3)Serum Th1/Th2 level(4)thyroid histopathology.Results:1.EAT model evaluation: The serum TG-Ab and TPO-Ab levels in model group were significantly higher than those in normal group at the 57 th day(P<0.05).2.Changes of thyroid weight index: There was significant difference between model group and normal group(P<0.05);there was significant difference between treatment group and model group(P<0.05);there was significant difference between Ruanjian Xiaoying Decoction group and Jinkui Shenqi Pill group(P<0.05);there was no significant difference between Ruanjian Xiaoying Decoction group and Jinkui Shenqi Pill group(P>0.05);and there was no significant difference between Ruanjian Xiaoying Decoction group and Jinkui Shenqi Pill group(P>0.05).3.Changes of thyroid hormone levels: FT3 and FT4 levels in model group were lower than those in normal group,TSH levels were higher than those in normal group,and there were significant differences between treatment group and model group(P<0.05);Ruanjian Xiaoying Decoction group and Jinkui Shenqi Pill were compared with Tripterygium wilfordii group(P<0.05);Ruanjian Xiaoying Decoction group and Jinkui Shenqi Pill were no significant differences compared with Jinkui Shenqi Pill group(P<0.05).Sex difference(P>0.05).4.Changes of serum TG-Ab and TPO-Ab: The serum levels of TG-Ab and TPO-Ab in model group were significantly higher than those in normal group(P<0.05);there were significant differences between treatment group and model group(P<0.05);there were significant differences between Ruanjian Xiaoying Decoction group and Jinkui Shenqi Pill group(P<0.05);there was no significant difference between Ruanjian Xiaoying Decoction group and Jinkui Shenqi Pill group(P<0.05);and there was no significant difference between Ruanjian Xiaoying Decoction group and Jinkui Shenqi Pill group.Sex difference(P>0.05).5.Changes of serum Th1/Th2 levels: Th1/Th2 in model group was significantly different from that in normal group(P<0.05);there were significant differences between treatment group and model group(P<0.05);there were significant differences between Ruanjian Xiaoying Decoction group and Jinkui Shenqi pill and Tripterygium wilfordii group(P<0.05);there was no significant difference between Ruanjian Xiaoying Decoction group and Jinkui Shenqi pill(P>0.05).6.Pathological changes of thyroid tissue: Compared with the normal group,a large number of thyroid follicular epithelial cells were destroyed and lymphocyte infiltration was obvious in the model group.Compared with the model group,the Ruanjian Xiaoying Decoction grouphad less destruction of follicular cavity and a small amount of lymphocyte infiltration,which was milder than the model group,and the recovery of follicular epithelial cells was better.Conclusion:1.Ruanjian Xiaoying Decoction has the effect of reducing lymphocyte infiltration in thyroid tissue.2.Ruanjian Xiaoying Decoction can inhibit hypothyroidism.3.Ruanjian Xiaoying Decoction can treat EAT by effectively reducing serum TG-Ab and TPO-Ab.4.Ruanjian Xiaoying Decoction can treat EAT by regulating Th1/Th2 balance.
