Experimental Study On The Clinical Effect Of Weikang No. 3 On Functional Dyspepsia Of Liver Depression And Spleen Deficiency And Its Effect On Brain-gut Peptide In Model Mice

Objective:1.Discussion the effect of Weikang Ⅲ in patients with functional dyspep-sia(FD)on clinical symptoms,signs and serum MTL and GAS of the effects.2.1n order to investigate the effects of Weikang Ⅲ on the expression of serum MTL,GAS,NT,CGRP,NT and CGRP receptors in the stomach,duodenum and brain of mice with functional dyspepsia.3.Effect the clinical effect and mechanism of Weikang Ⅲon patients with liver depression and spleen deficiency.Methods:l.Clinical study:65 patients,who will be included in the criteria of liver and spleen deficiency type FD,were randomly divided into Weikang Ⅲ group(the observation group)33 cases,the domperidone group(the control group)32 cases,the control group were treated with Weikang Ⅲ,and the control group was given the treatment of morphine tablets.After 2 weeks of treatment,observing the clinical symptoms,signs and serum MTL and GAS indexes.and optioning the 35 cases healthy volunteers in 22-63 year old.2.Experimental Study:80 clean Kunming mice weighing 18-22g,male and female haif,randomly were divided into normal group(30),model group(50).The unpredictable chronic stress model was established by food deprivation,wet cage feeding and other stimuli methods,observing the general of diet,hair and body weight of mice,including gastric emptying and small intestine propul-sion,the contents of MTL and GAS in blood serum,and pathological examination of gastrointestinal tissue of mouse for evaluation model.After the model succeed was randomly divided into three groups,were treated with distilled wa-ter,domperidone,Weikang Ⅲ-After 7 days,the contents change of GAS,MTL,NT and CGRP receptor in serum was tested by ELISA.The receptor expressions of NT and CGRP in antrum,duodenum and brain tissue were measured by immunohistochemical staining.Results:Clinical Research:1.The effective rate of gastric emptying in the observation group and the control group was 72.7%and 65.6%respectively.the observation group was better than the control group,between the two groups was not statistically significant on the differ-ence of gastric emptying(p>0.05).2.The total effective rate of the observation group were 81.8%and the control group were 74.9%,the observation group was superior to the control group,but the total ef-fecti-ve rate between the two groups was not statistically significant(p>0.05).3.Between the two groups in the levels of contents GAS and MTL was no significant difference(p>0.05),and the two groups between before and after were significant difference(p<0.01).Experimental Study:1.With the unpredictable chronic stress,the mice diet and drinking water are to low-er,slow growth of body weight,decrease the gastric emptying rate and small intestinal propulsion rate,the contents of MTL and GAS in serum are to lower,compared with the normal group was statistically significant,while the heantrum and duodenum tis-sue has not obviously pathology change.2.The model group the levels of contents MTL and GAS in the serum lower than those in the normal group(p<0.01),NT and CGRP in the serum higher than those in the normal group(p<0.01).Weikang Ⅲ group and domperidone group the levels of contents MTL and GAS in the serum of level rise,NT and CGRP in the serum of level low,compared with the FD model group was statistically significant(p<0.01 or p<0.05).But the difference between the two groups was not statistically significant(p>0.05)3.The model group of immune positive result of NT and CGRP receptor in the antrum and duodenum tissue were significant than those in the normal group(p<0.01).Weikang Ⅲ group and domperidone group of immune positive result of NT and CGRP receptor in the antrum and duodenum tissue were significant less than those in the FD model group(p<0.01),But the difference between the two groups was not statistically significant(p>0.05)4.The model group of immune positive result of NT and CGRP receptor in the brain tissue was significant than those in the normal group(p<0.01)Weikang III group and domperidone group of immune positive result of NT and CGRP receptor in the brain tissue was significant less than those in FD model group(p<0.01),But the difference between the two groups was not statistically significant(p>0.05).Conclusion:Clinical Research:This clinical trial shows that Weikang Ⅲ can improve the symptoms of patients and improve the quality of life,the mechanism may be through the regulation of serum MTL,GAS level to achieve.Experimental Study:1.With the unpredictable chronic stress can make the mice gastrointestinal function successfully building FD.2.Weikang Ⅲ can make the abnormal contents of MTL,GAS,NT,CGRP in serum significantly improving,which indicate that it therapy FD through regulating the level changes of brain-gut petide.3.Weikang Ⅲ can regulate the receptor expression of NT and CGRP in an-trum,duodenum,brain tissue,which is one of possible mechanism its treatment of FD.4.Combined with clinical and experimental research,based on the theory of "liver and spleen",it is speculated that Weikang Ⅲ treatment FD is through brain-intestinal in-teraction to achieve.
