Objective:This subject is to use the basic theory of TCM,the use of letrozole building,aromatase inhibitors from rat body weight,sex hormone level six,ovarian morphology,thermal shock immunoglobulin HSP27,HSP47,HSP90 expression rate in rat ovary and so on four aspects to analyze the wang yi by the side of kidney empty phlegmy wet polycystic ovary syndrome model rats,the mechanism and curative effect,to the wang yi fang treatment kidney phlegmy wet provide scientific theoretical basis for polycystic ovary syndrome.Methods:1.The group will take random number method selected 60 female level 6 weeks clean SD rats was divided into six groups,each group of ten,divided into normal group,model group,the British-35,three Chinese medicine group(wang yi by the high dose group,middle dose group,low dose group).2.The building and the treatment during the period of building,in addition to the normal group and 10,the rest of the group using letrozole building aromatase inhibitors,continuous lavage for 21 days.During the treatment,in addition to the normal saline lavage group and model group,control group adopts taie-35,three groups of Chinese traditional medicine group according to provide dose with the wang yi,continuous lavage for 15 days.Timing measurement during each rat body weight,feeding condition and record.3.After the specimen collection and test method of abdominal aorta take blood from detection between groups of rats serum T,LH,FSH,LH/FSH,E2,PRL and P ratio;Take bilateral ovarian tissue weigh each rat ovary weight,and calculate the volume;Put a free method to detect each group rats ovary HSP27,HSP47,HSP90 expression rate level;HE staining and put free method to detect each rat ovarian morphology and heat shock protein HSP27,47,90 image analysis.Results:1.The weight of rats Weight of rats in the six groups,the model group rats weight increase,the largest growth of normal rats weight minimum,the rest of the four groups all have different range of growth,but not more than model group.2.The serum sex hormone levels in rats2.1 T levels: normal group of rats serum T lowest,highest model group rats serum T.The treatment group compared with model group,P < 0.05,significant difference.Taie-35 after drug treatment group and the Chinese medicine group,lower serum T different range,high dose group was most pronounced in traditional Chinese medicine.LH highest 2.2 LH level: model group,compared with normal group(P < 0.05,the difference significant,building success.The treatment group compared with model group,P < 0.05,significant difference.Taie herbal medicines-35 group and treatment group,there was no statistically significant difference(P > 0.05,shows that the treatment group can reduce LH,in traditional Chinese medicine(TCM)was most evident effect of high dose group.2.3 FSH levels: normal group of rats serum FSH,highest model group compared with normal group,P < 0.05,the model of rat serum FSH,lowest building success.The treatment group compared with model group,P < 0.05,significant difference.Taie-35 with various traditional Chinese medicine group(P > 0.05,no significant difference.LH/FSH ratio level 2.4: model group LH/FSH ratio is the highest,compared with normal group,P < 0.05,significant difference.The treatment group compared with model group,P< 0.05,significant difference.Compared to the traditional Chinese medicine group and taie-35,all P < 0.05,with statistical significance.In Chinese traditional medicine best high dose group.3.Ria method to detect the rat ovaries of HSP27,HSP47,HSP90 expression rate3.1 in PCOS rat ovarian tissue,the expression of HSP27 level was lower than those of the other five groups of ovarian tissue of rats.3.2 in PCOS rat ovarian tissue,the expression of HSP90 level was lower than those of the other five groups of ovarian tissue of rats.3.3 in PCOS rat ovarian tissue,HSP47 expression level of high than other five groups of ovarian tissue of rats.4.Heat shock protein ovarian morphology 4.1 HSP27: high expression in normal group rats mature ovarian follicle oocytes and original follicular oocytes.4.2 HSP90: high expression in normal group of rat ovarian mature follicle oocytes,mesenchymal cells and granular cells.4.3 HSP47: lower expression in normal group rats ovaries of mature follicle mesenchymal cells.Conclusion:The wang yi is proved by adjusting the endocrine system in rats,decrease the serum T,LH,LH/FSH values,and by improving the body's immune heat shock protein HSP27/90/47 expression rate in the rat ovary in animal experiments have certain curative effect.The experiment for the wang yi by the clinical treatment of kidney empty phlegmy wet polycystic ovary syndrome provides scientific theoretical basis,as one of the effective clinical experience in the party.
