Objective: Obesity is the body's intake of heat greater than consumption,and excess heat accumulation in the body in the form of fat.Obesity can cause a variety of complications,such as T2 DM,dyslipidemia,polycystic ovary syndrome,sleep apnea syndrome and cancer.The acupuncture treatment of obesity has the advantages of easy operation,non-toxic side effects and significant curative effect.However,the mechanism of acupuncture treatment for obesity has not yet been fully elucidated.The purpose of this study is to explore the relationship between the release of inflammatory factors and the effect of mTOR protein on obesity by the acupuncture treatment.Methods: In this study,SPF grade SD male rats were taken as the experimental subjects,and the obesity rats were induced by high fat diet.After the obese model successful made,they were divided into model group,acupuncture group and Western medicine group according to random number table.Acupuncture group through acupuncture intervention,choosing the obesity rats acupoints,tianshu,guanyuan,Zusanli,fenglong,yinlingquan and other acupuncture points;The western medicine group was treated with metformin.The treatment time of western medicine group is same with the acupuncture group.During the treatment,the rat body weight,body length,lee 's index,blood sugar were inspected regularly.After 30 d intervention,the blood glucose and other indicators were measured.Then tissues,such as serum,hypothalamus and liver were taken to detect IL-6,TNF-alpha,IL-1 beta and MPO expression in serum by ELISA;The expression of mTOR protein in liver tissues of rats in different groups was detected by qrt-pcr and Western Blot.Analysis of the correlation between the expression of inflammatory factors IL-6,TNF-a,IL-1 beta,MPO and liver mTOR protein in serum of obese rats.Results:(1)After the treatment,the weight and blood glucose of the acupuncture group decreased significantly after the success of the model,with significant difference(P<0.05);(2)The expression of IL-6,TNF-a,IL-1 beta,MPO and m TOR protein in the model group was significantly higher than that in the blank group,with significant difference(P<0.05);(3)The expression of Il-6,TNF-,IL-1,MPO and m TOR in acupuncture group was significantly different from that in the model group(P<0.05);(4)The expression of mTOR protein in the liver of acupuncture group was decreased after acupuncture treatment,and there was significant difference between the model group after treatment(P<0.05).Conclusion: After acupuncture treatment,the weight and blood glucose of acupuncture group were significantly decreased,indicating that acupuncture has a good effect in reducing weight and reducing blood glucose.It is found that acupuncture treatment in obesity is related to the release of inflammatory factors and the expression of mTOR protein.
