Objective:This paper aims to observe the effects of electro-acupuncture(EA) at ST36 and ST45 treating high-fat diet-induced obese mice and influence on the gene expression of adiposetissueIL-6,TNF-a and MCP-1, as the experimental evidence of acupuncture to lose weight.Method:We choose 50 healthy and clean weaned C57BL/6J male mice to Adaptive feeding one week,and then randomly divide them into normal group and high-fat group, the normal group with normal diet, high-fat group with high fat diet, so as to establish food borne obese mice. After successful modeling, the obese mice are randomly divided into EA group and model group. Treatment methods:EA group are tied in daily home made holder to electro-acupuncture atST36 and ST45 of alternating side every other day, and the normal group and model group are only tied,observing and recording the weight and other indicators of each group of mice during treatment. After treatment, get serum specimens and fat, using real-time PCR to detect the expression ofadiposeIL-6, TNF-a, MCP-1, comparing the differences in mice weight,fat, blood sugar, lipid and other indicators of each group. Compare the fat morphological differences between groups of mice with HE staining.Result:EA significantly improves weight of obese mice:the weight of obese mice is significantly higher than normal group before treatment(p<0.01),and after treatment, EA group’s weight and weight loss rate is significantly reduced(p<0.01).EA significantly improves fat of obese mice:The visceral fat of fat-diet-induced obese mice is significantly higher than normal group(p<0.01), and the rate of fat and weight is also increased(p<0.01).After treatment, the EA group’s visceral fat is significantly decreased compared with the model group(p<0.01), and the rate of fatand weight is also decreased significant(p<0.01).EA significantly improves blood sugar, lipid levels of obese mice:The glucose, triglyceride,total cholesterol,Low-density lipoprotein in blood of obese mice are significantly higher than normal group.After treatment, the EA group’s glucose, total cholesterol and Low-density lipoprotein are significantly decreased compared with the model group(p<0.01).EA significantly improves the expression of adipose tissue IL-6-mRNA, TNF-a-mRNA, MCP-1-mRNAof obese mice:the expression of adipose tissue IL-6-mRNA, TNF-a-mRNA, MCP-1-mRNA of obese mice are significantly higher than normal group.After treatment, the EA group’s the expression of adipose tissue IL-6-mRNA, TNF-a-mRNA, MCP-1-mRNA are significantly decreased compared with the model group.Conclusion:Electro-acupuncture atST36 and ST45 can reduce weight, fat and blood sugar, lipid levels in mice,and inhibit the expression of adipose tissue IL-6-mRNA, TNF-a-mRNA, MCP-1-mRNA, thereby restraining differentiation and generation of adipocyte, promoting lipolysis. This method is good weight loss to food borne obese mice.
