Acupuncture Regulates PPAR-γ To Inhibit The Mechanism Of Aortic Inflammatory Response In Hypertensive And Obese Rats

Purpose : In recent years,the incidence of hypertension with obesity in China is increasing year by year,and there is a positive correlation between obesity and blood pressure.Therefore,by observing the effects of acupuncture on PPAR-γ,NF-k B signaling pathway,IL-6,ET-1,ICAM-1,VEGF and adiponectin and leptin responsible for insulin sensitivity in rats with hypertension and obesity,this study explains the efficacy of acupuncture on "Zusanli" and "Sanyinjiao" in reducing weight and lowering blood pressure.And the mechanism of protecting vascular inflammation injury by regulating PPAR-γ and its influence on NF-k B signaling pathway in hypertensive rats with obesity.Material and method:A total of 50 rats were selected,including 10 WKY rats and 40 SHR rats.In the blank group,10 WKY rats were selected,and the remaining 40 SHR rats were randomly divided into 4 groups: model group,obesity model group,acupuncture group and non meridian non acupoint group.After grouping,rats were fed with normal diet for one week.After one week,rats in blank group and model group were fed with normal diet,and rats in other groups were intervened with high-fat diet to prepare obesity hypertension model.Formula of high fat feed: 15% animal oil,18% sucrose and 3% egg yolk were added into the basic feed,which was sterilized and stored in a cool place.During the modeling period,blood pressure and body weight were measured twice a week and the data were recorded.When the weight of rats in the model group was more than 20% of that in the blank group,it was considered that the obese hypertensive rat model was successfully reproduced.After the animal model was successfully reproduced,the high-fat diet was stopped and the conventional diet was changed.After fixing the rats,the acupuncture group was intervened by acupuncture,and the acupoints were Zusanli and Sanyinjiao.The posterolateral part of the knee joint and 5mm below the head of fibula were selected at Zusanli for direct needling of 7mm;the internal malleolus tip of hind limb was selected at Sanyinjiao for direct needling of 5mm;the tail of rat was selected as the acupuncture control point in the non meridian and non acupoint group.After acupuncturing acupoints,Huatuo brand electroacupuncture instrument was connected,and the wave form was sparse and dense wave,in which the sparse wave was 1Hz and the dense wave was 5 Hz;the time of sparse wave was 10 s and the time of dense wave was 15 s,and the current was adjusted to the degree that slight convulsions could be seen in the local muscles of acupunctured acupoints and the rats kept quiet.Once a day,20 minutes for each treatment,6 consecutive days as a course of treatment and rest for 1 day,the treatment lasted for two weeks.The blank group and the model group were fixed with the same method as the acupuncture group for 20 minutes without any other intervention.After 4 weeks of treatment,all rats were deprived of food and water for 12 hours.After anesthesia,all rats were injected with chloral hydrate intraperitoneally.After anesthesia,the abdominal aorta was cut open with sterile surgical instruments to find out the abdominal aorta.The blood was taken out with a blood sampling needle.The adherent serosa around the aorta was removed.The thoracic aorta tissue was separated with a glass needle,and the fat in the terminal branch and surrounding tissue was pruned and placed in a cryopreservation tube Then put it in the refrigerator at-80℃ for storage.In addition,part of the aortic tissue was fixed in 4% paraformaldehyde for HE staining,and the fixed tissue was placed in a cool place for testing.The contents of leptin and adiponectin were detected by ELISA,the expression of PPAR-γ protein was detected by Western blot,the m RNA expressions of PPAR-γ,NF-k B,ET-1,ICAM-1,IL-6 and VEGF were detected by fluorescent quantitative PCR,and the morphological changes of aorta were observed by HE staining.Results:1.Acupuncture can reduce the systolic blood pressure and body weight of obese hypertensive rats.The experimental results showed that the weight and systolic blood pressure of obese hypertensive rats decreased significantly during acupuncture treatment,but the change of non meridian non acupoint group was not obvious,which indicated that acupuncture had significant effect on reducing weight and reducing blood pressure.2.Acupuncture can significantly increase the contents of leptin and adiponectin in rats.The expressions of leptin and adiponectin in the aorta of the model group and the obesity model group were significantly decreased(P < 0.05);compared with the model group,the expressions of leptin and adiponectin in the aorta of the obesity model group were significantly decreased(P < 0.05);compared with the obesity model group,the expressions of leptin and adiponectin in the aorta of the acupuncture group were significantly increased(P < 0.05)Compared with the acupuncture group,the content of leptin in the non meridian non acupoint group decreased,while the content of adiponectin increased,which was not consistent with the treatment trend of the acupuncture group.3.Acupuncture can increase the content of PPAR-γ protein in obese hypertensive rats.The content of PPAR-γ protein in the aorta of the model group and the obesity model group was significantly decreased(P < 0.05);compared with the model group,the content of PPAR-γ protein in the aorta of the obesity model group was significantly decreased(P < 0.05);compared with the obesity model group,the content of PPAR-γ protein in the aorta of the acupuncture group was significantly increased;compared with the obesity model group,the content of PPAR-γ protein in the aorta of the obesity model group was significantly increased Compared with the control group,the PPAR-γ protein content in the aorta of the non acupoint group did not change significantly(P > 0.05).4.Acupuncture can change the m RNA of PPAR-γ and its influence on NF-k B signaling pathway,and affect the m RNA expression of aortic inflammatory factors.The results showed that compared with the blank group,the m RNA expression of PPAR-γ in the aorta of rats in model group was significantly decreased,and the m RNA expression of NF-k B,ICAM-1,IL-6,VEGF and ET-1 were significantly increased(P<0.05);Compared with the model group,the m RNA expression of PPAR-γ in the aorta of rats in the obesity model group was significantly decreased,while the m RNA expression of NF-k B,ICAM-1,IL-6,VEGF and ET-1were significantly increased(P<0.05);Compared with the obesity model group,the m RNA expression of PPAR-γ in aorta of rats in acupuncture group was significantly increased,and the m RNA expression of NF-k B,ICAM-1,IL-6,VEGF,and ET-1 were significantly decreased(P<0.05).Conclusion:1.Hypertension can cause aortic inflammatory injury.After hypertension combined with obesity,the abnormal increase of body weight and systolic blood pressure will aggravate the vascular endothelial inflammatory reaction.2.Electroacupuncture intervention at Zusanli and Sanyinjiao can reduce the systolic blood pressure and body weight of obese hypertensive rats,improve the smooth degree of aortic intima,and reduce the vascular injury of obese hypertensive rats.3.Electroacupuncture intervention of Zusanli and Sanyinjiao can protect vascular inflammation by regulating PPAR-γ and NF-k B signaling pathway in hypertensive and obese rats.
