| Fenugreek(Trigonella foenum-graecum L)is an annual plant for medicine and food,the seed of fenugreek(fenugreek seeds)is an important traditional Chinese medicine.It has the effect of lowering blood sugar,lowering blood fat,reducing inflammation and relieving pain,warming kidney and Reinforcing Yang,and anticancer.Fenugreek seeds have a special aroma,it can be used as spices,or can be used as a coffee substitute after grinding.In this paper,under the subcritical condition,butane was used to remove the oil from fenugreek seeds,and study the crude polysaccharides and total flavonoids in fenugreek seed meal,it is hoped that it will provide theoretical basis and data support for the research and utilization of polysaccharides and flavonoids in fenugreek seeds,the main conclusions are as follows.A subcritical water extraction method was used to extract the crude polysaccharide in fenugreek seed meal.The extraction process was optimized by orthogonal technique and the influence of various factors was analyzed.The results showed that the influence of each factor on the extraction rate: time > liquid material ratio > temperature,the optimum extraction conditions of crude polysaccharide were selected and the highest extraction rate was 22.69%.A small amount of protein contained in the polysaccharide solution was found.The results showed that the protein had a gain effect on the emulsifying and emulsifying stability of polysaccharide,and the emulsifying property of polysaccharide was influenced by the concentration of polysaccharide,the test temperature,the solution pH and the ionic concentration in the solution.The total flavonoids in fenugreek seed meal were extracted with subcritical alcohol for the first time,and the extraction process was optimized by response surface method.The results showed that the ethanol concentration and extraction time had significant influence on the extraction rate.The optimum conditions were as follows: ethanol concentration 89.23%,ratio of liquid to material 62.73:1,extraction time 30 min,extraction temperature 143.1 ℃,The optimum extraction rate is 2.491%.The antioxidant activity of total flavonoids extracted by subcritical alcohol was tested in vitro.The results showed that the biological activity of flavonoids was not significantly affected,and the antioxidant activity in vitro was higher,and the scavenging rate of free radicals was affected by the concentration of flavonoids. |
