Phellinus is a rare Traditional Chinese Medicine,contains a variety of active chemicalcomponents such as polysaccharide and flavonoids,which biological activity has good anti-tumor activity anti-tumo and regulation of the immune.Flavonoids are polyphenoliccompounds,distribute in fruits, seeds and leaves,are an active ingredient of herbs.In thisexperiment,phellinus is the test material,used the total flavonoids as the marker,by the effect ofsingle factor test and response surface experiments to investigated alcohol extraction,ultrasonicand microwave assisted three extraction methods;to determine the optimum parameters of theextraction and compare three methods;through seven antioxidant system to research antioxidantof phellinus extract and refined flavonoids. The main results are as follows:1. The qualitative detecte of the five different leach liquor of Phellinus,through the colorreaction by specific reagents.The results of experimental show that leach liquor of Phellinuscontains flavonoids.2. Use colorimetric method to determine the content of total flavonoids:obtain a standardcurve equation A=0.122C+0.0086(R2=0.9997), the range of0-100ug/mL with good linearity inthe concentration.3. Useing the extract merhod of ethanol leach the Phellinus,through single factor experimentand response surface optimization analysis to obtain the optimal parameters:extractiontemperature67℃,75%ethanol concentration,material ratio of1:42and extraction5h. Under theabove tests conditions and measured total flavonoids extract content is26.91mg/g.4. Useing the extract merhod of ultrasonic assisted leach Phellinus, through single factorexperiment and response surface optimization analysis to obtain the optimal parameters r:ultrasonic power200W,acoustic processing time22min,material ratio1:33,68%ethanolconcentration.Under the above tests conditions and measured total flavonoids extract content is27.94mg/g.5. Useing the extract merhod of microwave assisted leach Phellinus,through single factorexperiment and response surface optimization analysis to obtain the optimalparameters:microwave time62s,microwave power550W,ethanol concentration70%,extractiontime2.2h.Under the above tests conditions and measured total flavonoids extract content is28.66mg/g. 6.Through analysis and comparison of the three test methods,we known thatmicrowave-assisted extraction greatly reduces the extraction time, increasing the total amountof flavonoids extracted. Determine microwave assisted extraction method is the best method.7.The seven different systems in vitro antioxidant tests showed that Phelligrins have someantioxidant activity and enhanced with increasing concentration: at reducing power system,theethanol extract and flavonoids refining capacity reduction of80%or more;the superoxide anionsystem,the scavenging effect of ethanol extracts,purified flavonoids and Vc to reach the samelevel, the clearperformance was good,and ethanol extracts of both clear and Vc maximum forceof about90%ance rate of80%:the hydroxyl radical system and DPPH· systems, various liquidscavenging;in nitrite system, refining82%scavenging flavonoids,followed by ethanol extract:R·clearing system, each scavenging of alkyl extracts not significant, up to60%; in H202system,refining the highest scavenging flavonoids, can reach78%, slightly higher than Vc scavenging.
