| Procapra Przewalskii(Procapra PrzewalskII)is a rare and endangered species endemic to China.Historically,it was distributed in Inner Mongolia,Gansu,Ningxia and Qinghai in China,and now it is only distributed around Qinghai Lake.Therefore,it is of great significance to apply modern molecular biology techniques to individual identification,population number estimation,genetic relationship identification and population viability assessment of Przewalskii.With the increase of population and the expansion of pastoral areas,human beings bring more and more pressure to the survival of Przewalski’s gazelle,leading to a great threat to the survival of przewalski’s gazelle and the decline of its population.Therefore,further protection and research of Przewalski’s gazelle are urgently needed.At present,the research on przewalskii mainly focuses on population protection,genetic diversity and breeding,but there are few reports on intestinal flora of wild animals.This study analyzed the feeding habits of Przewali in Ganzihe township and Hargai Town,and concluded that gramineae,legumes and compositae were the main plant groups in the grass green period.The plant with the largest daily feed on splendens and splendens and the least splendens is artemisia splendens.From the point of selectivity index,Praia splendens eats plants such as sacherophyta,likes to eat other plants such as Sacherella splendens,can eat plants such as rhizophora splendens and anorexanthus splendens.In this paper,the molecular composition and diversity of 15 intestinal microflora collected from Ganzihe Township and Hargai Town,Haiyan County,Qinghai Province were analyzed using high-throughput sequencing technology.Based on 97%sequence similarity,these sequences can be divided into 46 operational taxonomic units(OTU),with a total of 187 sequences,accounting for 55.2% of the total number of clones.Similarity to the 16 SrRNA gene sequence of known strains in Gen Bank and RDP II databases Less than 97%,representing unclassified,unidentified new strains.Phylogenetic analysis showed that the gut flora of the gazelle of Przewalski’s gazelle belonged to 4 phylums,in order: Firmicutes,Bacteroidetes,Actinobacteria and deformation Proteobacteria.The Firmicutes phylum includes more than half ofthe OTU and is the most diverse group of gut flora in the feces of Przewalski’s gazelle.Established a non-damaging Przewalski’s gazelle conservation genetics research technique,obtained the predominant flora present in the gut of gazelle,supplemented with basic data on gut flora of herbivores,and helped to reveal the gazelle The plateau adaptation mechanism provides a new idea for the protection of rare and endangered species. |
