Isolation And Identification Of Feline Panleukopenia Virus Strain And Preliminary Evaluation Of The Preparation,Test,Treat And Control Effects Of IgG F?ab?'₂

Feline panleucopenia?FPL?is an acute,potent infectious disease caused by Feline parvovirus virus?FPV?.It is one of the main infectious disease pathogens causing felines and the young cats are most susceptible to infection.Because of its rapid transmission and wide spread,it has brought serious economic losses to the pet cat and wild cat industry.FPV can cause severe diarrhea and hematochezia in animals,often accompanied by leukopenia,intestinal villus degeneration,high infectivity,high morbidity and mortality.With the widespread use of FPV vaccines in recent years,the incidence of FPV in cities has decreased significantly.However,the incidence rate in wild cats and big cats in captivity somewhat elevated.At present,there is no effective drug for direct treatment of FPV at home and abroad,and adjuvant therapy is often used to correct its dehydration,ensure electrolyte balance and prevent secondary bacterial infection.Because there are individual differences in animals,there is no guarantee that all sick animals will recover.The problem at this stage is to develop effective intestinal colonization treatment drugs to reduce mortality and reduce losses.In this study,an anal swab of a three month old kitten suspected of FPV infection was collected from the Animal Hospital of Jilin Agricultural University,and one virus was isolated by using F81 cells.Through morphological observation,hemagglutination and hemagglutination inhibition test,PCR identification,direct immunofluorescence test,whole gene sequencing and analysis,the isolated virus was identified as feline parvovirus virus.In order to obtain high-titer specific IgG F?ab?'₂ of feline parvovirus,we used cats as immune animals to prepare high-immune serum.The serum titer of the immunized cat serum was monitored by ELISA and serum was collected when the titers tended to be stable.According to the relevant provisions of the"People's Republic of China Pharmacopoeia"sterility test and safety inspection are in line with the requirements.The prepared F?ab?'₂ is applied to an animal emergency prevention and therapeutic test,and the body temperature,body weight,clinical symptoms and the anal swab of the animal are collected every day to detect the amount of detoxification.The comprehensive test results confirmed that the feline parvovirus-specific antibody F?ab?'₂ prepared in this study can alleviate the clinical symptoms of the affected animals,prolong the survival time and provide experimental data for the development of the feline parvovirus-specific antibody F?ab?'₂.
