This study was studied on obtaining a new pattern preparation of completing exogenous calcium,which is safe and high efficiency.It to be the superior substitute for the present preparation of enriching calcium by P.eryngii.The main results as following:(1)Analysis of effect exogenous calcium on growth and development of P.eryngii mycelium: Effects of five different exogenous calcium(calcium chloride,calcium nitrate,calcium carbonate,calcium lactate and calcium amino acid chelate)on P.eryngii mycelium were investigated.The calcium lactate was optimum exogenous calcium and the tolerance concentration was from 0.3 to 0.6mg/mL.In the growth of mycelium respect,the exogenous calcium concentration was 0.6 mg/mL,the fastest mycelium growth rate was showed.The exogenous calcium inhibited mycelium growth with 1.2 mg/mL.The mycelium growth decreased with the increase of the exogenous calcium concentration at a range of 0.6-2.4 mg/mL.In the fermentation broth with exogenous calcium,exogenous calcium for the amino acid chelate calcium showed that P.eryngii fermentation broth had the highest density.according to above results,the calcium lactate and amino acid chelate calcium were the most appropriate exogenous calcium.After fermentation,the calcium concentration of mycelium was significantly different.The highest calcium concentration in mycelium was 27.98 mg/kg with adding exogenous calcium lactate(0.6 mg/mL).(2)Analysis of effect exogenous calcium on growth and development fruiting body of P.eryngii: the level of total soluble sugar and soluble protein in fruit body was raised and then declined under treatment with different concentrations of calcium lactate.The effect of treatment group(0.6 mg/mL)was significantly higher than the control group(without calcium lactate).In the cultivated medium with different concentrations of exogenous calcium,calcium concentration was increased and then decreased,The effect of treatment group(0.6 mg/mL)was significantly higher than the control group.In the cultivated material,calcium concentration was increased by adding exogenous calcium,then the calcium concentration was declined in the fruiting body.Added the amino acid chelate calcium had similar effect as that of calcium lactate.Both optimum concentration of calcium lactate and amino acid chelate calcium in the cultivated material were 0.6 mg/mL.The calcium concentration of the medium was increased slowly by adding calcium lactate at 0.6 mg/mL,the highest calcium concentration of the P.eryngii fruiting body was 12.4 mg/kg.However,the calcium concentration of the culture medium increased slowly by adding calcium lactate and amino acid chelate calcium at 0.6 mg/mL,while the highest calcium concentration of the P.eryngii fruiting body was 9.4 mg/kg.In addition,the optimum exogenous calcium was able to improve the yield of P.eryngii fruiting body and short its growth cycle.The results showed that the yield and growth of P.eryngii had raised by adding low concentration of calcium lactate.It had supplied as a fundamental to developing new kind of organic calcium.
