| Edible mushrooms are not only delicious and nutritious,but also have medicinal values.In recent years,mushroom industry has developed rapidly.In 2016,the production value of China’s edible fungus industry has reached 274.178 billion yuan,and the output of the mushroom accounts for 75% of the world’s total production.As a multisystem metabolic disease,diabetes is complicated and difficult to cure completely,and can easily induce a series of complications,which seriously threatens the lives of patients.Polysaccharide as a natural ingredient can not only effectively relieve diabetes and its complications,but also does no harm to the body.It has become a hot spot for scholars all over the world.In this study,the industrialized cultivar Pleurotus eryngii was selected as the main research object,and the growth rate and yield of the mycelium were used as the main indicators to optimize the matrix formulation of Pleurotus eryngii and provide reference for improving the yield and quality of Pleurotus eryngii.Pleurotus eryngii fruit body polysaccharide(Pe-FBP)was extracted with water and precipitated with alcohol.Animal experiments were used to explore the role of Pleurotus eryngii polysaccharides in the treatment of diabetes,which would provide basis for the development and research of the new type of hypoglycemic and reducing lipid drugs.The results of the study are as follows:(1)Statistical analysis were made on the mycelial growth rate,average yield per bag,and biological efficiency of 45 groups of cultivation substrate formulations.It was found that formula 24(main ingredients: 23.33% of bran,19.6% of wood chips,46.00% of corn cob)was superior to other groups,in which,the growth rate of was 0.28 cm/d,the average yield per bag was 469.40 g,and the biological efficiency was 93.88%.(2)The effect of Pe-FBP on blood glucose in mice,the results showed that after gavage treatment for 4 weeks,the intervention of Pe-FBP(600mg/kg)can significantly decrease fasting blood glucose(FBG)and insulin resistance index(IRI)in mice compared with the model control group,which indicated that polysaccharides of fruit bodies of Pleurotus eryngiican play an auxiliary role in the treatment of diabetes to some extent.(3)Obese and streptozotocin(STZ)-induced diabetic mice exhibited dyslipidemia.The test found that Pe-FBP could reduce total cholesterol(TC),triacylglycerols(TG),and low-density lipoprotein cholesterol(LDL-C)levels,and increase high-density lipoprotein cholesterol(HDL-C)levels in mice,indicating Pe-FBP can improve lipid metabolism in mice.(4)The effect of Pe-FBP could increase the activities of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),and catalase(CAT),and decrease the content of malondialdehyde(MDA),indicating that Pe-FBP can protect pancreas from oxidative damage.(5)Inflammation test in mice showed that the Pe-FBP could reduce serum C-reactive protein(CRP),interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)levels after gavage,indicitaing that the polysaccharides have an inhibitory effect on diabetes-induced inflammatory responses to some extent.In summary,Pe-FBP can improve blood glucose and lipid metabolism disorders,and reduce oxidative stress injury to inhibit the progression of diabetic complications in obese and STZ-induced diabetic mice. |
