Pleurotus eryngii var.tuoliensis C.J.Mou and Pleurotus eryngii are two large groups of edible fungi.They are genetically close-related and both possess significant nutritious and economic values.In this case,an efficient and optimized cultivation method is practically important for the breeding of dominant strains.In this study,monokaryotic mycelium of 5 Pleurotus eryngii and 5 Pleurotus eryngii var.tuoliensis C.J.Mou were produced by using single spore separation technology.The single spore isolates were hybridized according to their mating type genes,and subsequently screened by using ITS-RFLP technology.The morphological comparison in different growth stages between two types of fungi under different temperature settings were carried out.Results showed clear distinct temperature effects on Pleurotus eryngii var.tuoliensis C.J.Mou and Pleurotus eryngii which are:(1)Six specific A factors and seven specific B factors were found in single spore isolates from 5 Pleurotus eryngii and 5 Pleurotus eryngii var.tuoliensis C.J.Mou strains accounting for 67% and 74% of the tested strains respectively.(2)3 purified hybrid strains were finally achieved from 21 treated strains by antagonism checking along with ITS-RFLP technology.(3)The growth rates of Mycelium of Pleurotus eryngii var.tuoliensis C.J.Mou and Pleurotus eryngii were positively correlated with the temperature changes.The optimum temperature for mycelial growth was discovered at 22°C.The growth rate of mycelium of two kinds strains were clearly different under this temperature,for instance,hybrid strain 11 D,53D.Moreover,the growth rate of mycelium of hybrid strain 70 D was different with its parental strain XB130 DB,but same with BL130 LA.(4)The primordium formation time of Pleurotus eryngii var.tuoliensis C.J.Mou is susceptible at the temperature interval of 4℃-24℃.The time of primordium formation was shortest while the temperature is in the range of 12℃-14℃.The primordium formation time of Pleurotus eryngii was negatively correlated with temperature.As the temperature increased,the primordium formation time became shorter,and it was shortest while the temperature is in the range of 22℃-24℃.The primordium formation time of the three hybrid strains 11 D,53D and 70 D was significantly different from that of the corresponding parent Pleurotus eryngii var.tuoliensis C.J.Mou at the statistical 0.05 level.(5)The shortest growth period for Pleurotus eryngii var.tuoliensis C.J.Mou fruitbody took place under the temperature of 13±1℃,while for Pleurotus eryngii,which was 23±1℃.The formation periods of the 3 hybrid strains were significantly different from that of the corresponding parental Pleurotus eryngii var.tuoliensis C.J.Mou at the statistical 0.05 level.(6)The stipe length of Pleurotus eryngii var.tuoliensis C.J.Mou is susceptible at the temperature interval of 4℃-24℃.The stipe length was longest while the temperature is at 12℃.The stipe length of Pleurotus eryngii was positively correlated with temperature.As the temperature increased,the stipe length became longer,and it was longest while the temperature is at 24℃.The stipe length of the three hybrid strains 11 D,53D and 70 D was similar to the stipe length of parental Pleurotus eryngii at the temperature interval of 4℃-24℃.(7)The cap diameter of Pleurotus eryngii var.tuoliensis C.J.Mou is susceptible at the temperature interval of 4℃-24℃.The cap diameter was longest while the temperature is at 12℃.The cap diameter of Pleurotus eryngii was positively correlated with temperature.As the temperature increased,the cap diameter became longer,and it was longest while the temperature is at 24℃.The cap diameter of the three hybrid strains 11 D,53D and 70 D was similar to the cap diameter of parental Pleurotus eryngii at the temperature interval of 4℃-24℃. |
