In this paper,through the systematic research on key techniques of Wasabi breeding,this paper seeks an effective solution to the problems of seed source degradation,low resistance and carrier virus in Wasabi planting.Through the research of the key technology of the series breeding,it can promote the standardization of the planting of Wasabi area,enrich the entire industrial chain of Wasabi,and then go international.The dormancy characteristics,seed viability and seed germination conditions of Wasabi seeds were studied in the study of Wasabi seed breeding.The effects of Wasabi pericarp extract on seed germination and seedling growth of Wasabi were studied.The effects of different soaking temperature,different hormone treatment and different proportion of soil on seed germination of Wasabi were studied.The strong respiration effect of Wasabi seeds was proved,and the soil needed should have certain air permeability.The total nutrient soil was found: the germination rate of the perlite = 1:2 combination reached 87.5 %;The activity of Wasabi seeds under different immersion water temperature,immersion time and cleaning interval time were studied.Immersion time & GT;The cleaning interval time was obtained and the optimal combination was obtained: that is,leaching 30 h at 15 °C and cleaning every 4.5 H;The effects of depericarp washing time,gibberellin soaking time,gibberellin soaking temperature and low temperature treatment time on seed germination of Wasabi were studied by orthogonal design experiment.Depericarp rinsing time & GT;Low temperature treatment time & GT;The best germination conditions were obtained by soaking the temperature of the amycin: after washing the 9d fruit by running water to remove the peel,the full seed was taken,and the erythromycin solution with a concentration of 75 mg/mL was soaked and treated with 4d at 5 °C.After treating 2d at a low temperature of 5 °C,sowing in the soil for 30 days,the germination rate of Wasabi seeds was as high as 97.92 %.In the study of Wasabi culture and propagation technique,the tissue induction,proliferation,plant regeneration and genetic transformation of Wasabi plant material were carried out.The induction of callus under different disinfection time,medium formula and culture temperature was studied when the true leaves,cotyledon,cotyledon and cotyledon of Wasabi seedlings were exosomes,and the best scheme was obtained: the cotyledon was exosomes.After 5 minutes of disinfection,then inoculated on MS +2,4-D1mg/L +6-BA 0.5 mg/L medium,culture at 20 °C,25d;The culture of secondary proliferation of different callus was investigated,and it was found that callus produced by exosomes was more easily proliferated,and callus was closely textured and non-browning,with a maximum multiplier of 2.48 ± 0.93.The adventitious Bud induction and adventitious root induction were investigated in different hormone ratio conditions,and the best culture formula for adventitious Bud differentiation was 1/2 MS + TDZ2 mg/L + IAA 0.5 mg/L.The best culture medium formula for rooting induction was 1/2 MS + NAA 0.1 mg/L + IBA 0.05 mg/L + AC 0.1 mg/L;The aseptic acquisition of Wasabi seeds,the activation culture of genetically transformed strains,the genetic transformation of different implants and the proliferation of Wasabi hairy roots were studied,and the optimal conditions for genetic transformation of Wasabi plants were as follows: wasabi leaves were planted.Through the pre-culture process,36 d culture with the bacteria can obtain a higher genetic conversion rate.
