Bletilla striata(Thund.)Reichb.f.is a perennial herb of Rhizoma Bletillae genera,which is also known as ZiLan.Owing to its excellent effect and remarkable economic value,thus received extensive attention of many scholars.The demand of the market doubled every year with the in-depth study of Bletilla striata,Caused people crazyly excavated Rhizoma Bletillae wild resources in recent years.Which leads to its wild resources decreased sharply.For now the wild resources of Bletillae had been unable to meet the rapid development of medicine industry demand.Therefore artificial cultivation of herbs Bletillae striata has become an unavoidable topic.How to reasonable close planting and achieve harvest in the autumn has become a key problem of planting Bletilla striata standardly.Recently scholar both at home and abroad mainly focus on its extracting valuable components or pharmacological activities,but little work has been done in breeding area.We did a lot of researchs on the technology of direct seeding,rapid multiplication,microbe fertilizer to Bletilla striata.Finally summed up a set of rapid propagation technology,and provided people with a safe,rapid,reliable production technology route.The findings are as following:(1)In the research of inducting Bletilla seed germination.The type of culture medium?6-BA?NAA were sleeted as the main factors of inquiry research,which the seeds germination medium was optimized through orthogonal test at last?Now we know the best seeding medium composition was MS+6-BA2.0mg/L+ NAA1.0mg/L.In this condition Seed germination rate can reached 91.8%.The analysis results from the orthogonal range shows that the factors of influence on seed germination can be sorted by size:The type of culture medium>6-BA>NAA.The result of Bletilla striata rooting culture test reveal Banana juice can increase the formation of adventitious root and NAA could promote the proliferation of leaves.After evaluations the best rooting medium was MS+Banana juice(70g/L)+NAA(0.5mg/L).(2)In the experiment of transplanting matrix,the rate of plantlets survival reached 82%,when it grows on fertile soil.And the rate of plantlets survival reached 70.4%.Reflect the native soil has promoting effects of microorganisms on plant.In the lawsuit,we speculate that the sand drain to substrate and can increase the permeability of soil,the fermented bark containing inorganic nutrients which can be easily absorbed,while provides a microbial symbiosis environment for plants(3)Through the observation of Bletilla striata and phenology,breeding system studies and determination of testing seed vigor in the seedling experiment.We conclude that the seed of Bletilla striata belongs to the type easy germination.We finally chose breeding method:Petri dish + Gauze + adorable(humus soil:sand:bark shavings,=4:1:2).Considering the characteristics of seed resources is huge,direct seeding is undoubtedly a new seedling of high efficiency,it has practical significance for the development and utilization of plant resources.(4)Through the microstructure of Bletilla striata mycorrhizal observation,found that root structure of mycorrhizal fungi community is single,hyphae are distributed in root cortex cells and the formation of mycelia.Mycelia of adjacent cells could be conected through hyphae,which was twisted like ropes.Through the comparison between the hand sectioning,acid lactic acid fuchsin stained squash method,aniline blue staining method of pressing the three microscopic method,we concluded that the image is clear,the background color of smaller interference,mycelial characteristics particularly obvious in microscope observation.In addition,this method had many advantages,for exemple its doesn't need special equipment,simple operation,less time-consuming,make it be widely used in the preliminary observation of Orchidaceae plants mycorrhizal structure.(5)Through in situ experiment,we successfully induced Bletilla striata seed germination and Isolated several kinds of fungi by the seedling cup.After morphological and molecular identification,the findings are that these 26 species belonging to 12 genera,which mainly contain:Chaetomium aureum,Rhizoctonia,Fusarium oxysporum,Colletotrichum,Neosartorya fennelliae etal.(6)In the screening test of the fungus.The plantlets in vitro carried in the inoculation and incubation experiments with mycorrhiza fungi.Statistics the biomass of plants.In the end,picking out the fungis which has the better promoting effect.These mycorrhiza fungi are as follow:14,19,w-1-1,s-20,16,s-15-2.After the identification of the strains,2 strains belong to Rhizoctonia,3 strains belong to Chaetomium,l strains belonged to Penicillium.In these six kinds of mycorrhizal.It indicated that Mycorrhizal fungi in root are concentrated in Rhizoctonia and Chaetomium.(7)In the experiment of promoting mechanism of mycorrhizal fungi.Through using modern biological technology to investigate the ability of fungi.The results showed that the tested 6 strains of PGPR could produce plant hormones IAA and ABA in varying degrees.Besides some fungus in addition to the above features,also has the properties of soluble phosphorus,ammonia.Although the result maybe not reflect the truth of mechanism,but it can be the auxiliary supply mycorrhizal fungi promoting the growth of plant substances in a way.
