In recent years,as people pay more attention to the quality and safety of livestock products,Chinese herbal medicine additives as antibiotic substitutes and nutritional regulators have become a research hotspot.As an important part of natural vegetation in Ningxia,sophora alopecurosa is a plant of leguminous and sophora genus,with a wide distribution of wild resources.Due to its high medicinal value,it has been listed as one of the six authentic medicinal materials under special protection in Ningxia province.The medical value of sophora alopecurolobata has been developed in anticancer,antibacterial immunity,blood pressure reduction,antitoxin and other functions.Tan lamb as experimental animal in Ningxia,this paper using in vitro fermentation and animal feeding experimental research method of combining,adding different levels in the diet is studied to Tan lamb in vitro fermentation of sophora alopecuroides,growth performance,nutrient digestion and blood physiological and biochemical indexes,slaughtering and meat quality,as Chinese herbal medicine additive in feed for sophora alopecuroides provide theoretical basis for scientific applications.Effects of different levels of alopecuroides alopecuroides on gas production,fermentation parameters and volatile fatty acids were studied by rumen fermentation in vitro.Seven male Tan sheep with good body condition and similar body weight(31.42±1.05 kg)were selected for fattening,and were raised as tumor gastric juice donors in a single cage in a self-made metabolic cage.The five treatment groups were supplemented with 0%,0.25%,0.5%,1%,and 2%of the diet respectively.Three replicates were designed for each treatment group,and the in vitro gas production culture for 72h and the in vitro fennenter culture experiment for 48h were carried out continuously.The results are as follows:With the increase of adding amount of sophora alopecuroides,acid washing fiber degradation rate(ADFD)presents the downward trend,adding amount was 1%and 2%in significantly lower than that of control group(P<0.05),the adding amount was 0.5%,1%,2%,significantly reduced when the methane yield(P<0.05),the increase of adding amount was 0.5%and 1%hydrogen production(P<0.05),adding different levels of sophora alopecuroides on 72 h gas production and other gas yield no significant influence(P>0.05),the nutrient solution pH value,NH3-N no significant influence(P>0.05),The addition of sophora alopecuroides significantly reduced the content of volatile fatty acids(P>0.05).Combined with the results of in vitro fermentation,it was found that the addition of sophora alopecuroides inhibited the degradation ability of acid washing fiber,and then affected the generation of VFA in rumen fermentation,resulting in the reduction of methane emissions.To study the effects of different levels of sophora alopecuroides on growth,digestion,rumen fermentation parameters,blood biochemical index,slaughter and meat quality of Tan sheep.50 castrated male beach sheep of 6 months old with similar body weight(28.02±1.32 kg)and similar body condition were selected.The sheep were divided into 5 groups using the single-factor completely randomized design.The 5 treatment groups were added with 0%,0.25%,0.5%,1%and 2%of the diet respectively.10 sheep were in each test group for 60 days,including 10 days of pre-feeding and 50 days of trial.The results showed that:(1)When adding 2%bitters to the diet,the average daily intake,average daily weight gain and feed weight ratio of Tan sheep were significantly increased(P<0.05).The addition of sophora alopecuroides had no significant effect on slaughter performance(P>0.05).(2)The addition of sophora alopecuroides had no significant effect on the digestibility of various nutrients(P>0.05).(3)The addition of bitter beans to the diet reduced the concentration of NH3-N in the rumen(P<0.05)and the concentration of is butyric acid(P<0.05).(4)Dietary addition of 2%bitter beans significantly increased the concentration of total protein(TP)in serum(P<0.05).When the dosage was 1%and 2%,the blood glucose concentration was significantly reduced(P<0.05).Cholesterol levels in the four groups were lower than those in the control group,but the difference was not significant(P>0.05).Other blood biochemical indexes had no significant effect(P>0.05).(5)In terms of meat quality,meat color A value at the addition level of 0.5%was significantly higher than that of the control group(P<0.05).The addition level of 0.5%and 1%significantly increased the water loss rate of the longest dorsal muscle(P<0.05).Addition levels of 0.5%,1%and 2%significantly reduced the cooked meat rate of the longest dorsal muscle(P<0.05).The addition level of 1%significantly increased the initial water content of the longest dorsal muscle(P<0.05).The intramuscular fat content of each experimental group was significantly lower than that of the control group(P<0.05),and the change trend was a downward trend with the increase of the amount of bitter beans added(6)dietary addition of bitter beans had no significant effect on the muscle inosine and inosine in tan sheep(P>0.05).(7)Add sophora alopecuroides level was 0.5%,1%and 2%,significantly lower back muscle longest total fatty acid content and saturated fatty acid content(P<0.05),had no significant effect on single unsaturated fatty acids(P>0.05),significantly increased when adding level of 2%polyunsaturated fatty acid and n-6 functional fatty acid content(P<0.05),add a level of 0.5%,1%and 2%,significantly increase the ratio of PUFA/SFA(P<0.05).(8)Adding level was 2%,significantly increased the longest back muscle,lysine and the content of phenylalanine,each group back muscle in the longest total amino acid content was increased by 1.93%,3.91%,3.90%and 9.64%respectively,each group must the amino acid was increased by 4.82%,6.39%,8.21%and 16.56%respectively,add to the flavor of sophora alopecuroides amino acids have no significant difference(P>0.05).To sum up,the addition of sophora alopecuroides into the diet inhibited the methane emission of beachsheep and improved the growth performance of beach sheep.Improved meat quality,The optimum dietary addition of sophora alopecuroides was 2%.
