Protective Effect Of Adsorbents On The Toxic Effects Of Zearalenone On Sow Uterus | | Posted on:2020-10-11 | Degree:Master | Type:Thesis | | Country:China | Candidate:W B Gao | Full Text:PDF | | GTID:2393330572997216 | Subject:Basic veterinary science | | Abstract/Summary: | | | Zearalenone(ZEA)is an estrogenic mycotoxin produced by Fusarium spp.and ZEA can interfere with animal reproductive function in many ways.Many studies have confirmed that pigs are extremely sensitive to ZEA.Excessive intake of ZEA can affect the growth and digestion and absorption of piglets and sows,and cause abnormal reproductive organs development and sow reproductive dysfunction.In order to reduce the intake of ZEA and reduce the adverse effects of ZEA on pig production,it is necessary to detoxification of pig feed by ZEA.At present,adding ZEA adsorbent to feed is one of the most widely used physical detoxification methods of feed ZEA.In this study,a new composite adsorbent provided by the Feed Research Institute of the Chinese Academy of Agricultural Sciences was used to verify and analyze its effective protective effect on uterine toxicity of ZEA in reserve sows at different doses.48 hybridized reserve sows were randomly divided into 6 groups(n=8).The control group was fed with basal diet,zearalenone group was fed with basal diet + 1.0 mg/kg ZEA,the three dosage groups were fed with basal diet + 1.0 mg/kg ZEA + different dosages(1.0,2.5,5.0 g/kg),and the adsorbent control group was fed with basal diet + 1.0 mg/kg ZEA + 2.0mg/kg.5g/kg montmorillonite.Pre-test for 7 days,28 days.The protective effect of the new adsorbent on uterine toxicity of zearalenone-induced reserve sows was evaluated by measuring the production performance,estrogen content in serum,serum protein and some enzymatic activities of experimental animals,and analyzing the expression of proliferating cell nuclear antigen(PCNA),ghrelin,heat shock protein 70(Hsp70)and autophagic factor LC3 in uterus,which provided detailed information for the popularization and application of adsorbent.Real theoretical basis.The results showed that: 1.Three doses of the new adsorbent had no significant effect on the average daily gain and feed-Weight ratio of the reserve sows,but 0.1% and 0.5% doses of the new adsorbent and montmorillonite could significantly reduce the swelling of the vulvaand the area of the vulva.2.New adsorbent had no significant effect on serum protein of reserve sows,and montmorillonite reduced IgG content;there was no significant difference in serum alkaline phosphatase and creatine kinase content among groups;0.25% and 0.5%dosage of new adsorbent and montmorillonite significantly increased glutamic oxalate transaminase content;0.1% and 0.5% dosage of new adsorbent and montmorillonite group had no significant change in Glutamic acid dehydrogenase content,0.25% dosage of new adsor The dosage of 0.5% new adsorbent and montmorillonite significantly reduced the increase of estrogen in serum caused by ZEA.3.Histological observation showed that the three doses of new adsorbents could effectively improve the increase of uterine glands caused by ZEA and the abnormal proliferation of uterine glands.Among them,0.5% group had the best effect,but the effect of montmorillonite was not obvious.4.The expression of PCNA protein in 0.25% and 0.5% dose groups was significantly lower than that in ZEA group.The expression of PCNA in 0.5% dose group was the closest to that in control group,and the number of PCNA-positive cells in uterine glands was significantly reduced.The new adsorbent significantly reduced the number of ghrelin-positive cells and protein expression in uterine glands,of which the 0.25% dose group had a significant reduction effect.The dosage reduced the number of Hsp70 positive cells in uterine cavity epithelium and glandular epithelium,and the effect was most obvious in 0.5% group,while the expression of LC3 was increased in 0.5% adsorbent group,and the highest expression was found in 0.5% adsorbent group.These results suggest that the new adsorbent can effectively reduce abnormal uterine proliferation and oxidative stress injury induced by ZEA.5.In montmorillonite group,the expression of PCNA,ghrelin and Hsp70 protein was lower than ZEA group,similar to 0.1%adsorbent group,and the expression of LC3 protein was higher than ZEA group.Conclusion: The results of this experiment indicate that the new adsorbent has an effective protective effect on reproductive toxicity of gilts in gilts.Comprehensive analysis of relevant test indicators,with the best effect of 0.25% of the amount of additives. | | Keywords/Search Tags: | gilts, zearalenone, uterus, adsorbent, PCNA, Ghrelin, Hsp70, LC3 | | Related items |
