Piglets’ diarrhea has also brought great economic losses to farmers while affecting the healthy development of the pig industry.Taurine has advantages such as safety,high efficiency,stable physical and chemical properties,and diversified functions,so it has been used as an additive for animal diets and has been widely used in animal production.Therefore,attention is increasingly paid to its production practice and clinical application.This study was to investigate the protective effect of taurine on intestinal mechanical barrier function of piglets induced by lipopolysaccharide and its mechanism.In this study,24 healthy piglets were selected and divided into 4 treatment groups according to the principle that the proportion of males to females accounted for half: blank control group(C),taurine group(T),lipopolysaccharide(M)group and taurine + lipopolysaccharide group(MT).6 replicates per treatment,one piglet per replicate.The control group and the lipopolysaccharide group were fed the basal diet,and the taurine group and the taurine+lipopolysaccharide group fed the diet with 0.3% taurine added to the basal diet.On the early morning of the 28 th day of the test period,the lipopolysaccharide group and the taurine + lipopolysaccharide group were injected intraperitoneally with 100 μg/kg·BW of lipopolysaccharide.The blank control group and the taurine group were injected with the same dose of 0.9% physiological body weight.brine.One hour after the injection of lipopolysaccharide or physiological saline,all piglets were fed D-xylose solution at a dose of 0.1 g/kg·BW.Four hours after the injection of lipopolysaccharide,blood was collected from the anterior vena cava and the serum was separated.Each group was then selected.Three pigs weighing close to each other were immediately slaughtered and samples of small intestine were taken.The results showed that:(1)The study of growth performance of piglets supplemented with 0.3% taurine in the diet found that compared with the control group,the ADG of the taurine group piglets increased significantly(P<0.05),and the difference between ADFI and F/G was not significant(P>0.05).(2)The study of the apparent digestibility of piglets supplemented with 0.3% taurine in the diet found that the metabolic rates of Ash and P in the taurine group were significantly higher than those in the control group(P<0.05).There was no significant difference in the metabolic rate of CP,EE and Ca(P>0.05).(3)The results of ELISA showed that: compared with the control group,the contents of DAO,ET,D-LA,IFABP,COR,and IL-1β in serum of group M increased significantly(P<0.01),and that of D-xylose The content was significantly reduced(P<0.01);Compared with group M,the serum levels of DAO,ET,D-LA,IFABP,COR and IL-1β were significantly decreased(P<0.05),and the content of D-xylose was extremely high in MT group.Significant increase(P<0.01),C group and T group compared the above indicators were not significant differences(P>0.05);thus shows that taurine can inhibit the immune response caused by LPS caused by inflammatory Excessive secretion of factors and increased intestinal permeability improve intestinal absorption.(4)Observing the results of paraffin section,compared with the control group,the VL,CD and VL/CD in the duodenum,jejunum and ileum of the T group were not significantly different(P>0.05),compared with the C group.The VL of the jejunum in the M group was significantly lower(P<0.05).Compared with the M group,the VL,VL/CD in the duodenum,jejunum,and ileum of the MT group increased to varying degrees(P>0.05,P<0.05).CD was significantly lower(P< 0.05).(5)Real-time fluorescence quantitative PCR assay showed that: compared with the control group,the relative expression levels of ZO-1,Claudin-1,Occludin mRNA in the duodenum,jejunum,ileum and colon of the T group were not significantly different.(P>0.05);Compared with the control group,the relative expression levels of ZO-1,Claudin-1,and Occludin mRNA in duodenum,jejunum,ileum and colon decreased in different degrees(P<0.01,P<0.05);Compared with M group,the relative expression levels of ZO-1,Claudin-1,Occludin mRNA in duodenum,jejunum,ileum,and colon of MT group increased to varying degrees(P<0.01,P<0.05);This indicates that taurine can significantly increase the expression of tight junction protein related genes in the intestinal mechanical barrier of piglets caused by LPS.In summary,dietary supplementation of taurine can significantly inhibit LPS-induced excessive release of pro-inflammatory cytokines in piglets,significantly improve intestinal mucosal structure of piglets,reduce intestinal permeability,and enhance the expression of tight junction proteins in intestinal mucosa.There by enhancing the intestinal mucosal mechanical barrier. |
